摘要
目的 :自行研制抗人雄激素受体 (androgenreceptor,AR)单克隆抗体 (McAb) ,以期达到可替代国外AR多克隆抗体。方法 :采用人雄激素受体合成肽 (hAR -N)交联钥孔戚血蓝素 (KLH)为抗原 ,免疫BALB/C小鼠 ,经用Kohler和Milstein杂交瘤技术成功地获得 4株能稳定分泌抗hAR单克隆抗体 (McAb)杂交瘤细胞株 5 5 ,141,2 0 7,2 5 7。结果 :对 45例前列腺癌组织石蜡切片进行了免疫组化 (IHC)检测 ,同时对自制抗hAR -NMcAb与进口抗AR多抗进行分析比较 ,阴阳总符合率为 91%。结论 :自制抗ARMcAb特异性强 ,灵敏度高 ,结果与Zymed公司产品相同 ,操作简便 ,适于临床应用。
Objective:To study and prepare anti human androgen receptor(AR) monoclonal antibodies (McAb).Methods:Four cell lines of hybridoma secreting specific McAb against AR were first established by fusion SP2/0 cells with spleen cells from BALB/C mice immunized with the coupling complex of hAR-KLH.Results:Paraffin-embedded sections of 45 prostate cancers were detected.There was an overall concordance of 91% using IHC between AR ployclonal antibody from Zymed and hAR-N McAb in our work.Conclusion:The results show that the McAb obtained in this study would be a useful tool to detect the AR status in prostate cancer.
