卷烟烟气暴露对小鼠肺巨噬细胞吞噬功能和调节性T细胞的影响 被引量:1

摘要 目的观察卷烟烟气对小鼠肺泡巨噬细胞(AM)的影响,探讨调节性T(Treg)细胞与AM吞噬功能的关系。方法 BALB/c小鼠经卷烟烟气暴露1、2、3个月后处死,姬姆萨染色计数支气管肺泡灌洗液(BALF)中AM数目,吞噬D大肠杆菌(E.coli)检测AM吞噬功能,流式细胞仪检测外周血及BALF中Treg细胞的比例,蛋白液体悬浮芯片技术检测BALF炎性细胞因子。结果小鼠暴露卷烟烟气1个月,BALF中AM数显著增高,AM吞噬功能和BALF中Treg细胞比例显著降低,炎性细胞因子白介素(IL)-1β、单核细胞趋化蛋白-1(MCP-1)和巨噬细胞炎症蛋白-1α(MIP-1α)含量增高,IL-10量减少;暴露2个月,AM数量及其吞噬功能和BALF中Treg细胞比例均显著降低,MCP-1及MIP-1α含量增高,IL-1β、IL-10含量减少;暴露3个月,除IL-10含量增高外,其他观察指标与对照组比较,差异无统计学意义。结论卷烟烟气对小鼠AM功能有影响,Treg细胞与AM功能关系密切,并受炎性细胞因子调节。 Objective To investigate the effect of cigarette smoke exposure on phagocytic function of alveolar macrophage(AM) and regulator T(Treg) cells in mice.Methods The BALB/c mice were executed on 1,2 and 3 month after exposure to cigarette smoke.The AM were collected to count and test phagocytosis functions by swallowing E.coli.The ratio of CD4+CD25+Foxp3+Treg cell in peripheral blood and bronchoalveolar lavage fluid(BALF) were analyzed by flow cytometry.The cytokines in BALF were examined by Bio-plex System.Results Compared with control group,1 month after cigarette smoke exposure,the number of AM and the concentration of IL-1β,MIP-1 and MCP-1α increased,and phagocytic function of AM,the ratio of BALF CD4+CD25+Foxp3+Treg cells in CD4+T cells and the concentration of IL-10 decreased.2 months after cigarette smoke exposure,the number and phagocytic function of AM,the ratio of BALF CD4+CD25+Foxp3+Treg cells in CD4+T cells and the concentration of interleukin(IL)-1β reduced,and the concentration of monocyte chemotactic protein-1(MIP-1) and macrophage inflammation protein-1α(MCP-1α) increased.However,3 months after cigarette smoke exposure,all indicators showed no statistical difference from normal group other than the concentration of IL-10(increased).Conclusion There are obvious effects of cigarette smoke exposure on phagocytic function of mice macrophage and Treg cell,and some inflammatory factors are probably concerned with the process.
