摘要
目的 揭示巨噬细胞集落刺激因 子(M-CSF)对单核/巨噬细胞氧化损伤的影响。方法采用真核细胞基因导入 的脂质体转染方法,建立了两株M-CSF基因过表达的RAW264.7细胞系,并观察 了氧化剂叔丁基氢过氧化物(tbOOH)对该RAW264.7细胞的氧化损伤作用。结果用M-CSF基因转染的RAW264.7细胞具 有较强的分泌有活性M-CSF的能力,而与正常RAW264.7细胞相比,M-CSF 过表达的RAW264.7细胞更易受到氧化剂的损伤。结论在一定条件下,M-CSF可促进巨噬细胞的氧化损伤。
Objective To investigate the effect of macrophage colony-stimulating factor(M-CSF)on the oxidative injury to monocytes/macrophages.MethodsTwo M-CSF overexpressed RAW264.7 cell lines were established by applying transfection met hod.The oxidative injurious effect of te rtbutyl hydroperoxide (tbOOH) on the est ablished cell lines was also investigate d.ResultsThe two M-CSF -transfected RAW264 .7 cell lines showed high capacity of se creting M-CSF.Comparing to the non-trans fected RAW264.7 cells,the M-CSF-overexpr essed RAW264.7 cells were more vulnerable to oxidative injury.ConclusionIn some occasions,M-CSF could aggravate the oxidative injury to macrop hages.
