HPLC测定血浆和尿液中头孢匹胺的浓度

Determination of cefpiramide in human serum and urine by HPLC

目的 建立头孢匹胺在人体血浆和尿液中的快速HPLC测定方法。方法 血浆和尿液样品经去蛋白、解离处理后 ,用RP HPLC进行分析。采用DiscoverODSC1 8柱 2 5 0mm× 4.6mm ,5 μm ,流动相为甲醇 三乙胺醋酸液 水 =6 40∶3∶135 7,pH 5 .38,紫外检测波长为 2 5 4nm ,乙酰苯胺为内标。结果 血浆和尿液中头孢匹胺回收率为 (99.5 8± 2 .94) %和 (98.72± 1.97) % ,在 1.2 5～ 16 0 μg·ml- 1 和 2 .5～ 16 0 μg·ml- 1 范围内线性良好 (r=0 .999) ,日内、日间RSD均小于 5 % ,重现性良好 ,RSD小于 7%。结论 本法简便、准确、灵敏、经济 。

OBJECTIVE To establish an RP HPLC method for the determination of cefpiramide(CPM) level in plasma and urine.METHODS After precipitating protein and extracting,the concentrations of CPM were analysed by RP HPLC.The separation was performed in a Discovery ODS C 18 column (250 mm×4.6 mm,5 μm) with a mobile phase of methanol:acetic triethylamine H 2O=640∶3∶1357(pH 5.38 ).The UV detector at 254 nm was selected.The internal standard was acetanilide.RESULT The recoveries of CPM in plasma and urine were (99.58±2.94)% and (98.72±1.97)%,respectively.The calibration curves were linear over the range of 1.25～160 μg·ml -1 in seurm and 2.5～160 μg·ml -1 in urine with a correlation coefficient of 0.999.The RSD was less than 7%.CONCLUSION The method is simple,precise,accurate and usable for studies in human.
