摘要
目的 :探讨肿瘤细胞在接受 γ射线照射后多核细胞的形成与细胞放射敏感性的关系。方法 :利用定时显微摄影装置对人膀胱癌细胞株 RT112和人胚胎肠上皮细胞株 I40 7在 8Gyγ射线照射后的细胞改变进行 2 40 h连续观察。结果 :两个细胞株受 γ射线照射后不仅出现细胞的凋亡和坏死 ,还有双核或多核细胞的形成。两个细胞株的双核或多核细胞的形成方式各不相同 ,I40 7主要是细胞融合 ,RT112则基本上是由于细胞分裂失败而形成 ,其双核或多核细胞的形成指数分别为 42 .9和5 4.7,除少数最终凋亡和坏死外 ,大部分持续生存直至观察结束 ,但由于未能进行有效的分裂 ,可视为增殖性死亡。而对照组细胞未发现双核或多核细胞的形成。结论 :实验结果证明不仅细胞凋亡和坏死影响着细胞的放射敏感性 ,多核细胞的形成也起着重要的作用。
Objective:To estimate the relationship between formation of multinucleate cell after exposure to 8Gy of gamma irradiation and radio sensitivity in tumor cells Methods:Two cell lines,RT112 from bladder cancer and I407 from normal embryonic intestinal epithelium,exposed to 8 Gy of gamma irradiation were observed during 240 hours by time lapse microscopy Results:Apoptosis and necrosis were detected in the cell lines In addition,formation of bio or multi nucleate cells was also found In I407 the formation was by fusion of cells and in RT112 by failure of cell division The indices of formation were 42 9 and 54 7 respectively Few of the bio or multi nucleate cells died by apoptosis and necrosis and most of them survived to the end of observation Since the cells survived did not undergo sufficient proliferation they were be identified as dead On contrary,no bio or multi nucleate cell was detected in control Conclusion:The results suggested that not only apoptosis and necrosis influenced radio sensitivity of these cell lines,but also the formation of bio or multi nucleate cells was one of the important factors
