摘要 建立一种 HPL C法分离和检测人血浆中吡柔比星的分析方法学。以柔红霉素为内标 ,1ml血浆加0 .1ml(0 .4mol/ L,p H=9.0 )的氯化铵缓冲液碱化后以 CHCl3∶ CH3OH(2∶ 1)混合溶剂提取 2次。在Phenomenex(C1 8)柱上 ,以 0 .0 4mol/ L KH2 PO4(p H=3.0 )∶ CH3CN=6 9∶ 32为流动相 ,荧光检测波长 480 /5 5 0 nm(Ex/ Em)。结果 ,血浆中吡柔比星的线性范围 5~ 10 0 0 μg/ L(r=0 .997) ,方法的回收率为 95 .3% ,日内及日间变异性 RSD均小于 5 %。结论 :本方法适用于临床吡柔比星的血药浓度监测和药代动力学研究。 To estabish a HPL C assay method for the determination of pirarubicin( PRB) in plasma. Daunorubicin( DNRB) was used as the internal standard.Plasma samples were alkalized by0 .4 mol/L NH4 Cl buffer( p H9.0 ) ,and extracted with CHCl3∶CH3OH( 2∶ 1) 2 times.Separation was carried outon a2 50 mm× 4 .6mm( 5μm) Phenomenex column and a mobile phase of0 .0 4 mol/L KH2 PO4 ( p H=3.0 )∶ CH3CN( 69∶ 32 ) . Fluorescent detector set at an excitation wavelength of4 80 nm with an emission wavelength of550 nm.The standard curves for the PRB is linear within the range of5~ 10 0 0 μg/L in plasma.The minimal absolute recoveries of PRB is over90 .0 % .The within day and day- to- day validations were less than5% .The method has been used in clinical therapeutic drug monitoring and pharmacokinetics study of PRB.
