摘要
目的 :建立一种敏感、特异和快速检测石蜡包埋组织中多肿瘤抑制基因 (MTS1 )的方法。方法 :利用改良的 DNA制备方法 ,成功的从甲醛固定、石蜡包埋组织中提取出 DNA,同时用建立的多重聚合酶链反应 (PCR)进行检测。结果 :30例石蜡包埋的正常组织与新鲜组织提取的 DNA经 PCR扩增后 ,电泳图谱都出现 5 36 bp和 310 bp带 ,即结果相同。结论 :只要提取、纯化得当 ,石蜡包埋组织在分子生物学研究领域中将有重要应用价值。
Objective:To establish a sensitive,specific and rapid method to detect MTS1 from formaldehyde fixed and paraffin embedded tissue Methods:We succeeded in extracting DNA from formaldehyde fixed and paraffin embedded tissue by reformed method of DNA preparation And we detected it by mutiplex PCR Results:There are similar results between 30 pairs of paraffin embedded tissue and fresh tissue of the normal Eletrophoresis maps of amplification products all preserved 536 bp and 310 bp band Conclution:It suggests that paraffin embedded tissue will be of value in molecular biology research if we extract and purify it properly
