摘要
目的:通过检测肝癌细胞系HepG2和HepG2.2.15细胞中基质金属蛋白酶-2和-9的表达水平及两种细胞的生长速度,探讨MMPs在HBV的影响下对肝细胞癌增殖能力的影响及其与肿瘤侵袭转移之间的关系。方法:用免疫组化法检测HepG2细胞及转染了HBV表达质粒的HepG2.2.15细胞中MMP-2和MMP-9的表达情况,用四甲基偶氮唑盐比色法(MTT法)检测细胞增殖情况并观察两种细胞的某些生物学活性。结果:MTT及细胞生长曲线显示HepG2细胞的生长速度快于HepG2.2.15细胞。免疫组化检测HepG2细胞中MMP-9的表达高于HepG2.2.15细胞,而MMP-2的表达在两种细胞中均不明显,无显著性差异。结论:通过体外检测肝细胞癌细胞系HepG2细胞和HepG2.2.15细胞生长状态的差异,揭示HBV对肝细胞癌的生长、增殖具有一定的抑制作用,HBV能够抑制肝细胞癌中MMP-9的表达,从而间接影响了其侵袭和转移的能力。
Objective: To investigate the effects of MMPs on the proliferation of hepatocellular carcinoma under the influence of HBV and to review the relationship between MMPs and tumor invasion and metastasis, by testing the expressions of MMP - 2 and MMP - 9 in the ceils of HepG2 and HepG2.2.15 and the speed rate of the cells in vitro. Methods : HepG2 cells and transfected HBV expression plasmid HepG2.2.15 cells were selected to test the expressions of MMP - 2 and MMP - 9 with immunohistochemical method, and the cell proliferation was detected with MTI" method to observe some biologic activities of the cells. Results : MTI" and the cell growth curve showed that the growth rate of HepG2 was faster than that of HepG2. 2.15. Immunohistochemical detection showed that the expres- sions of MMP -9 in HepG2 cells was higher than that in HepG2.2.15, however, there was no significant difference of the expressions of MMP - 2 between the two kinds of cells. Conclusion : It is revealed that HBV has some inhibi- tory action to the growth and proliferation of hepatocellular carcinoma by testing the difference of growth tendency be- tween the cells of HepG2 and HepG2.2.15 in the cell line of hepatocellular carcinoma. HBV can restrain the ex- pression of MMP - 9 in the cells of hepatocellular carcinoma so as to influence the ability of tumor invasion and me- tastasis indirectly.
