摘要
该文利用曲里苯蓝(Trypan blue)能与多糖形成复合物的原理,建立起一种简便、快捷筛选纤维素酶产生菌的改良方法。实验以羧甲基纤维素钠作为底物,配制0.02%曲里苯蓝培养基培养纤维素酶产生菌株,检测菌落周围形成的透明水解圈,水解圈的大小同酶活高低有一定指数关系。该方法与传统方法相比,不仅有着同样高的灵敏度,而且不影响菌株生长,可以应用于纤维素酶改良工程菌株的筛选。
A plate assay based on the formation of haoles on petri dishes, containing 0.02% trypan blue dye and polysaccharides as substrates, provides a specific, reliable and rapid detection of cellulose decomposition microbes. After one cultivation periods, the colonies of cellulose decompose mi- cro-organisms cultivated with CMC-Na were surrounded by some distinct zones on the medium. The size of hydroids had exponential relationship with the enzyme activity. Compared with traditional methods, it can provide high sensitivity, harmless to microorganisms, and also has potential of as an efficient method using in cellulase engineering techniques.
