生大黄、清半夏、常山对大鼠血浆AChE和脑-肠轴β-EP的影响研究

Effects of crude rhubarb,Rhizoma Pinelliae Preparata,and Dichroa febrifuga on plasma AChE activity and β-EP levels in brain-gut axis among normal rats

摘要 目的:探究生大黄、清半夏、常山对正常大鼠血浆乙酰胆碱酯酶(AChE)活力和脑-肠轴β-内啡肽(β-EP)含量的影响。方法:将32只健康Wisrar大鼠随机分为对照组、生大黄组、清半夏组和常山组,分别灌胃生理盐水、生大黄水煎液、清半夏水煎液和常山水煎液,连续10d。测定各组大鼠的血浆AChE活力和血浆、下丘脑、胃窦、小肠中β-EP含量。结果:生大黄和清半夏组血浆中AChE活力增大(P<0.01,P<0.05),常山组活力降低(P<0.05);生大黄组大鼠下丘脑、胃窦、小肠中β-EP含量降低明显(P<0.05,P<0.01),血浆中β-EP含量有上升的趋势;清半夏组大鼠胃窦、小肠中β-EP含量降低较明显(P<0.05),下丘脑和血浆中含量变化不大;常山组大鼠血浆中β-EP含量下降趋势较大(P<0.05),其他位点含量变化不明显(P>0.05)。结论:生大黄、清半夏和常山能明显影响AChE活力和β-EP的含量,对消化系统的调节具有重要的作用。 Objective: To investigate the effects of crude rhubarb,Rhizoma Pinelliae Preparata,and Dichroa febrifuga on plasma acetylcholinesterase(AChE) activity and β-endorphin(β-EP) levels in brain-gut axis among normal rats.Methods: Thirty-two healthy Wistar rats were randomly divided into control group,crude rhubarb group,Rhizoma Pinelliae Preparata group,and Dichroa febrifuga group to receive saline,water decoction of crude rhubarb,water decoction of Rhizoma Pinelliae Preparata,and water decoction of Dichroa febrifuga,respectively,by gavage for 10 consecutive days.Plasma AChE activity and β-EP levels in the plasma,hypothalamus,gastric antrum,and small intestine were measured.Results: The crude rhubarb group and Rhizoma Pinelliae Preparata group had significantly increased plasma AChE activity(P < 0.01 or P < 0.05),while the Dichroa febrifuga group had significantly decreased plasma AChE activity(P < 0.05).The crude rhubarb group had significantly decreased β-EP levels in the hypothalamus,gastric antrum,and small intestine(P < 0.01 or P < 0.05) and showed an increasing trend in plasma β-EP level.The Rhizoma Pinelliae Preparata group had significantly decreased β-EP levels in the gastric antrum and small intestine(P < 0.05),but with little changes in β-EP levels in the hypothalamus and plasma.The Dichroa febrifuga group showed a significant declining trend in plasma β-EP level(P < 0.05),but no significant changes in β-EP levels were seen in other sites(P > 0.05).Conclusion: Crude rhubarb,Rhizoma Pinelliae Preparata,and Dichroa febrifuga have marked effects on AChE activity and β-EP levels and play important roles in regulating the digestive system.
