摘要
目的 :研究非病毒载体法基因转染的最适条件及其潜在价值。方法 :利用脂质体介导和配体定向法将带有野生型 P5 3 基因 ( Wt- P5 3 ) c DNA的质粒引入细胞 ,用直接镜检、软琼脂培养、四甲基偶氮唑 ( MTT)法和乳酸脱氢酶法等研究细胞生长特性的改变情况。结果 :绝大多数细胞被转染 ,肿瘤细胞生长受到明显抑制而发生凋亡。结论 :本方法对于基因体外转染是可行的 ,并且有可能将来用于小细胞肺癌的 P5 3
Objective:To optimize the P 53 gene transfection conditions and value the nonviral gene vector methodMethods:The plasmids with wildtype P 53 gene (WtP 53 ) were liposomemediated,ligandtargeted,and introduced into cancer cells,then the comparative studies were made with direct microscopy,soft agar culture,monotetrazolium (MTT) analysis and lactate dehydrogenase (LDH) testResults:Most cells were transfected,the growth of which was apparently arrested while apoptosis was observedConclusion:The gene transfection protocol was practical in vitro and might be useful in P 53 gene therapy for small cell lung cancer
