胃旁路术后GK大鼠的铁代谢及血糖改变

摘要 目的:观察胃旁路术后2型糖尿病GK大鼠血清铁、铁蛋白、总铁结合力及血糖的改变。方法:将30只12周龄的雄性GK大鼠随机分为手术组、假手术饮食控制组及假手术空白对照组各10只,观察手术前后铁代谢、血糖、进食量和体重变化,并于术前和术后第4、8周行口服糖耐量实验。结果:术后8周,胃旁路术组空腹血糖较术前明显下降(P<0.01),糖耐量曲线下面积、血清铁和铁蛋白均明显降低(P<0.01),总铁结合力明显上升(P<0.01)。结论:2型糖尿病大鼠胃旁路术后血糖改善,血清铁、和铁蛋白显著下降,总铁结合力升高。 Objective To observe the changes of serum iron(SI) and ferritin(SF) and of total iron binding capacity(TIBC) in type-2 diabetic Goto-Kakizaki rats after gastric bypass surgery and to study the effect of iron metabolism changes on type-2 diabetes.Methods Thirty male GK rats were randomly divided into three groups :Group-O rats underwent gastric bypass surgery Group-C rats underwent sham operation and were pair-fed with group-O rats;Group-B rats underwent laparotomy and had free access to diet after operation.The blood glucose,food intake and body weight were recorded before and after surgery.The serum iron,serum ferritin and total iron binding capacity were determined before surgery and 4 weeks and 8 weeks after operation.Results In Group O,serum parameters before and 8 weeks after surgery were as follows :Serum glucose (17.6±2.1vs,8.2±1.1,P0.01),SI(215.18± 36.55 vs 86.65±26.61,P0.01),SF(1.076±0.273 vs 0.768±0.178,P0.01),and TIBC(455.40±81.96 vs 643.11± 58.77,P 0.01) In Group B and C,the parameters levels of iron metabolism were not different.Conclusion Gastric bypass surgery reduces serum iron concentration in type-2 diabetic rats and this effect may be involved in the improvement of blood glucose seen in the same rats.
