CIK/NK细胞对荷骨肉瘤NOD/SCID小鼠肺转移瘤的杀伤作用 被引量:2

Effect of cytokine-induced killer(CIK) cells and natural killer(NK) cells on pulmonary metastasis in U-2OS NOD/SCID mice

摘要 目的制备骨肉瘤U-2OS细胞NOD/SCID小鼠肺转移模型,探讨CIK/NK细胞对肺转移瘤的杀伤作用。方法 60只NOD/SCID小鼠随机分为3组(每组20只),A组:每只鼠经尾静脉接种U-2OS细胞2×106(0.2 ml)后24 h,输注CIK/NK细胞(2×107/0.2 ml);B组:每只鼠经尾静脉接种U-2OS细胞2×106(0.2 ml)后24 h,注射生理盐水0.2 ml;C组:每只鼠经尾静脉注射生理盐水0.2 ml后24 h,注射生理盐水0.2 ml。比较各组小鼠生存时间及肺部肿瘤负荷情况。结果 B组接种U-2OS细胞后29 d内小鼠全部死亡,其中16只肺部出现肉眼可见瘤块;A组接种U-2OS细胞后60 d内小鼠死亡8只,肺部均发现肉眼可见瘤块,余12只肺部均无瘤块。A组平均生存时间显著高于B组,差异有统计学意义(P<0.05),而与C组比较差异无统计学意义(P>0.05)。B组小鼠死亡时体重明显低于A组,而肿瘤细胞比例明显高于A组(P<0.05)。结论 CIK/NK细胞在NOD/SCID小鼠体内有肯定的抗骨肉瘤肺转移瘤效果。 Objective To study the effect of CIK/NK cells on pulmonary metastasis using NOD/SCID mice model for osteosarcoma pulmonary metastasis. Methods Sixty NOD/SCID mice were randomly divided into 3 groups. There were 20 mice in each group. Group A: each mouse was inoculated with 2x10^7 (0.2 ml) CIK/NK cells 24 hours after 2x10^6 (0.2ml) U- 2OS cells infused. Group B: each mouse was inoculated with 0.2 ml normal saline 24 hours after 2x10^6 (0.2 ml) U-2OS cells infused. Group C: each mouse was inoculated with 0.2 ml normal saline 24 hours after 0.2 ml normal saline infused. The survival time and the tumor cell load of the lung of each group were compared. Results All the mice of group B died within 29 days, tumors were visible in lung of 16 mice. Eight mice of group A were died within 60 days, tumors were visible in lung of the 8 mice. At the time of 60 days tumors were not visible in lung of the remaining 12 mice. The survival time of group A was significantly longer than that of group B (P 〈0.05). However, there were no significant difference in survival time between group A and C(P 〉0.05). The weight of group B was significantly lighter than A(P 〈0.05). The ratio of tumor cells of group A was significantly lower than group B (P 〈0.05). Conclusion CIK/NK cells have the certain effect of anti-osteosarcoma lung metastases in NOD/SCID mice.
