牛膝醇提物对急性软骨损伤兔软骨的影响

Effect of Alcohol Extract of Achyranthes on the Cartilage in Rabbits with Acute Cartilage Damage

目的观察牛膝醇提物体对急性软骨损伤兔软骨病理学的影响。方法将24只新西兰大白兔随机分为对照组、低剂量组、中剂量组、高剂量组各6只,采用锐器刻痕法进行急性软骨损伤模型造模后,低、中、高剂量组分别给予牛膝醇提物1、3、6g/(kg·d)灌胃,对照组给予等量蒸馏水,每日2次,共灌胃4周。采用免疫组织化学方法检测Ⅱ型胶原表达,并进行软骨HE染色和改良Mankin's评分。结果高、中、低剂量组Ⅱ型胶原表达阳性百分率明显高于对照组,改良Mankin's评分低于对照组(P<0.05或P<0.01);并且高剂量组Ⅱ型胶原表达阳性百分率亦高于中、低剂量组,改良Mankin's评分低于中、低剂量组(P<0.05);中剂量组Ⅱ型胶原表达阳性百分率高于低剂量组,改良Mankin's评分低于低剂量组(P<0.05)。结论牛膝醇提物能有效刺激急性软骨损伤兔软骨细胞增殖,提高Ⅱ型胶原表达,修复软骨损伤,并且呈剂量依赖性。

Objective To observe the effect of alcohol extract of Achyranthes （Radix Achyranthis Bidentatae） on the cartilage in rabbits with acute cartilage damage. Methods Twenty-four New Zealand white rabbits were randomized into the control group, low dose group, middle dose group and high-dose group, with 6 in each. The acute cartilage damage models were established with sharp indentation. The low-dose, middle-dose and high-dose groups were gavaged with 1, 3 and 6g/ （kg . d）alcohol extract of Achyranthes. The control group was given same amount of distilled water. The intragastric administration was given to each group twice daily for 4 weeks. The expression of type-II collagen was detected by immunohistochemistry. The cartilage HE staining and modified Mankin＇s score was observed. Results Comparing with the control group, the percentage of positive expression of type-II collagen was significantly increased but the modified Mankin＇s score was significantly decreased in the high-dose, middle-dose and low-dose groups （P〈0.05 or P〈0.01）, especially in the high-dose group （P〈0. 05）. Comparing with the low-dose group, the percentage of positive expression of type-II collagen was significantly increased but the modified Mankin＇s score was significantly decreased in the middle-dose group （ P 〈 0.05）. Conclusion The alcohol extract of Achyranthes can effectively stimulate chondrocyte proliferation, increase type-II collagen expression and repair cartilage damage with a dose-dependent manner.
