大黄丹参对胰腺纤维化大鼠氧化损伤修复作用的研究 被引量:3

摘要 目的观察大黄丹参对胰腺纤维化大鼠氧化损伤的修复作用。方法 50只雄性Wistar大鼠随机分为正常组,模型组,大黄丹参低、中、高剂量组,每组10只。采用腹腔注射DDC 6周建立胰腺纤维化模型。低、中、高剂量组在造模同时分别给予相应剂量的大黄丹参灌胃6周[分别为0.137 g/(kg.d),0.274 g/(kg.d)和0.548 g/(kg.d)],正常组和模型组予等量生理盐水灌胃,疗程结束后取材。HE染色观察胰腺病理学变化,酶标法及分光光度法测定大鼠血清及胰腺MDA、SOD、LDH及GSH含量。结果各剂量组大黄丹参均可不同程度改善胰腺大体形态学变化。三组治疗组血清和胰腺的SOD及GSH活力明显升高,与模型组比较有统计学差异(P<0.05,P<0.01),其中均以高剂量效果最显著(P<0.05,P<0.01),呈一定的剂量依赖性。各剂量组血清和胰腺的MDA含量均明显降低,与模型组比较具有统计学差异(P<0.05,P<0.01),其中血清中MDA含量变化以高剂量组最显著(P<0.01)。高剂量组可降低血清LDH含量,与模型组比较有统计学意义(P<0.01),各剂量组均可降低胰腺中LDH含量(P<0.05,P<0.01),其中以低剂量效果最显著(P<0.01)。结论大黄丹参可通过降低LDH及MDA含量、提高SOD及GSH活力,增强机体抗氧化能力,阻断氧化过程,抑制脂质过氧化对胰腺细胞长期的攻击,减轻胰腺损伤来缓解胰腺纤维化进程,并呈现一定的剂量依赖性。 Objective To explore the oxidative damage repair effect of Rhubarb and Salvia Miltiorrhiza on pancreatic fibrosis rats. Methods Fifty male Wistar rats were randomly divided into five groups with 10 respectively:normal group, model group, low - dose group, medium - dose group and high - dose group. To establish pancreatic fibrosis model by intraperitoneal inject DDC for 6 weeks. At the same time,the 3 treatment groups were given the corresponding drugs(0. 137g/kg · d-1,0. 274g/kg · d -l and 0. 548g/kg · d-1 respectively). Rats were sacrificed at the end of treatment, HE for pancreas histological changes, enzyme standard instrument law and spectrophotometric methods for the content of MDA and the activity of SOD, LDH and GSH in pancreatic tissue and serum. Results Different doses of drugs can improved the gross morphological changes of pancreas with different degrees. The activity of SOD and GSH in serum and pancreas in the 3 drug groups were increased obviously compared with model group and with a dose dependent( P 〈 0. 05, P 〈 0.01 ). The content of MDA was degraded markdly in pancreas and serum in the 3 drug groups compared with model group( P 〈 0. 05, P 〈 0.01 ), and in serum the high dose group was the most significant effect( P 〈 0. 01 ). The content of LDH in serum was degraded in the high dose group with a significant differences compared with model group (P 〈 0.01 ) ;At the same time,the content of LDH in pancreas was reduced obviously in the 3 different drug doses compared with model group(P 〈0. 05 ,P 〈0.01 ) ,and the low dose group has the most significant effect( P 〈 0.01 ). Conclusion Rhubarb and Salvia Miltiorrhiza can reduce the LDH and MDA content, improve the SOD and GSH vitality, enhance organism anti - oxidize ability, block oxidation process, restrain lipid peroxidation pancreatic cells to reduce pancreatic damage and ease the process of pancreatic fibrosis, and show a dose dependent.
