赤雹根总皂苷对类风湿性关节炎大鼠足跖组织中NF-κB p65活性作用的研究

Inhibitory Effect of Total Saponin of Thladiantha dubia on Express Level of NF - κB p65 in Paw tissue of rats with RA

目的研究赤雹根总皂苷(SRTD)对类风湿性关节炎大鼠足跖组织中NF-κB p65蛋白表达的影响,探讨SRTD治疗类风湿性关节炎的机制。方法雄性SD大鼠80只,随机取10大鼠作为正常对照,其余大鼠采用弗氏完全佐剂诱导佐剂性关节炎,将造摸成功的50只大鼠随机分为模型组,赤雹根总皂苷160,80,40 mg/(kg.d)组和雷公藤多苷12 mg/(kg.d)组,各组分别给予相应药物,模型组ig蒸馏水,连续21 d。计算各组大鼠关节炎指数,给药后第22天,处死大鼠。用免疫组化法和Western blot法检测足跖组织中NF-κB p65蛋白的表达。结果赤雹根总皂苷能明显降低RA大鼠的关节炎指数(P<0.01,P<0.05),显著下调NF-κB p65蛋白的表达(P<0.01,P<0.05)。结论 SRTD对大鼠类风湿性关节炎有明显治疗作用,下调NF-κB p65蛋白的表达是其作用机制之一。

Objective To investigate the effect of the total saponin of Thladiantha dubia（SRTD） on the NF- κB p65 protein expression in tissue of rheumatoid arthritis （RA） ratg paw. Methods 80 male SD rats were randomly taken 10 as control group, and the rest used freundg adjuvant induced adjuvant arthritis. 50 rats which successfully induced by adjuvant were randomly divided into five groups：SRTD 160 mg· kg-1 · d-l ,80 mg ·kg-1 · d 1,40 mg· kg-1 · d-mgroups and Tripterygium Glycosides 12 mg · kg-1· d-Jgroup. Each group was given corresponding medicine, and control group was given distilled water,intragastrie admin- istration for 21 days. The arthritic index of rats was calculated;all rats were killed at 22d. The expression of NF- κB p65 proteins in rats paw tissues was detected by immunohistoehemistry and western holt. Results Arthritic indexes of AA rats in STFD groups were lower than those in model group（ P 〈 0. 01 ,P 〈 0.05 ） , and the expression of NF - κB p65 protein was significantly decreased （P 〈0. 01 ,P 〈 0.05 ）. Conclusion The SRTD has therapeutic effect on RA rats, the decreased the expression of NF- κB p65 protein is one of the mechanisms.
