摘要
目的:探讨藜蒿黄酮提取物对人肝癌细胞SMMC7721凋亡及迁移的影响。方法:采用TUNEL法检测藜蒿黄酮提取物对细胞凋亡的诱导作用;Boyden趋化小室检测细胞迁移抑制作用;分光光度法检测caspase-3活性;免疫细胞化学法检测细胞P21的含量。结果:藜蒿黄酮提取物诱导SMMC7721细胞凋亡,抑制细胞迁移,伴随细胞内caspase-3活性及P21含量升高。结论:藜蒿黄酮提取物可诱导SMMC7721细胞凋亡并抑制其迁移,其机制与caspase-3和P21有关。
OBJECTIVE To investigate the effects of flavonoids extract of Artemisia selengensis Turcz in apoptosis and mi- gration of human hepatoma SMMC7721 cells. METHODS TUNEL assay was used to detect apoptosis induced by the fla- vonoids extract of Arternisia selengensis Turcz; the cell migration was evaluated using Boyden chamber assay; the caspase-3 ac- tivity was determined by spectrophotometric method; the p21 protein was determined by immunohistochemistry. RESULTS The flavonoids extract of Artemisiia selengensis Turcz could induce apoptosis and inhibit migration of SMMC7721 cells, accom- panied by elevated easpase-3 activity and P21 protein level. CONCLUSION The flavonoids extract of Artemisia selengensis Turcz can induce SMMC7721 cells apoptosis and inhibit migration, the mechanism may be involved caspase-3 and P21.
