稻瘟病菌假定几丁质酶家族基因的表达特点 被引量:4

摘要 在稻瘟病菌基因组数据库中检索到15个假定几丁质酶相关基因,借助生物信息学分析,发现这些几丁质酶均具有糖苷水解酶18家族结构域,有的还具有几丁质结合功能域;亚细胞定位分析表明,8个编码蛋白定位在细胞外,其余分别定位在细胞质和线粒体。通过与酵母几丁质酶CTS1与CTS2的聚类分析,发现这些假定几丁质酶可以分为2大类,其中11个假定几丁质酶与CTS2聚为一类,即CTS2同源蛋白;其余4个与CTS1聚为一类,即CTS1同源蛋白。在此基础上,本研究进一步通过Real-time PCR技术分析这些假定几丁质酶编码基因在稻瘟病菌菌丝生长、孢子形成、芽管萌发和附着孢形成阶段的表达模式。结果显示,除了MGG_01876.6,其他14个基因在稻瘟病菌不同生长发育阶段均有表达,说明几丁质酶家族蛋白可能参与稻瘟病菌的生长发育、形态建成和致病过程。 Genome-wide analysis of chitinase genes in the Magnaporthe oryzae genome database revealed 15 putative chitinase-related genes. They all contain the glycosyl hydrolases family 18 (Glyco 18) domain, while some of them also have the chitin binding site (ChtBD1) or carbohydrate binding site (CBS). Eight of them are predicted to be secreted, while others are predicted to be in the cytoplasm and mitochondria. Phylogenetic analysis showed that the 15 putative chitinases can be classified into two clusters. Eleven of them are homologs of CTS2 of Saccharomyces cerevisiae, while the other four genes are closely related to CTS1 of S. cerevisiae. Furthermore, Real-time quantitative PCR showed that all the chitinase-related genes except for MGG_01876.6 are expressed at all fungal development stages including mycelia, conidia, germ tubes, and appressoria formation. Our result indicated that chitinase-related genes may function in morphogenesis and pathogenesis of M. oryzae.
