摘要
目的:探索一种简便制备大鼠急性肺缺血再灌注损伤模型的方法,并对其效果进行评估。方法:将30只Wistar大鼠随机分成2组(缺血再灌注组和假手术组),每组15只。假手术组单纯开胸,缺血再灌注组采用显微无创血管夹夹闭左肺门60min,松开血管夹后再灌注120min,建立急性肺缺血再灌注模型。术后获取标本,检测肺湿干重比值,肉眼、光镜观察肺组织结构变化。结果:肺缺血再灌注组湿干重比值明显高于假手术组(P<0.01),肉眼、光镜下均可观察到肺损伤改变。结论:成功构建了Wistar大鼠急性肺缺血再灌注损伤动物模型,操作简单,成功率较高,为肺缺血再灌注损伤机制研究提供了一种可行的动物模型。
Objective:To prepare and evaluate a new type of acute lung I/R injury model in rats.Methods:Thirty Wistar rats were randomly divided into two groups: IR group and sham group.We establish a lung I/R injury model,the rats underwent left lung ischemia,after 60 minutes of ischemia,reperfusion for up to 2 hours was evaluated.The ratio of lung W/D weight was compared between two groups,and the changes in lung structures were observed under the visual inspection and light microscopy.Results:The rate of lung W/D weight in I/R group was significantly higher(P0.01)than in control group.Compared with IR group and sham group in light microscopy,there were more serious damages in IR group.Conclusion:It is simple and easy to establish a model of lung I/R injury in rats,maybe useful in studying the mechanism of lung I/R injury.
