正常SD大鼠心血管系统内皮素转换酶表达水平及意义

摘要 目的 :探讨正常 SD大鼠心脏及血管 (动脉、静脉 )内皮素转换酶 (Endothelin- converting EnzymeECE) m RNA的分布。方法 :半定量 RT- PCR法检测组织中的 ECE m RNA,同时以甘油醛 - 3-磷酸脱氢酶 (Glycer-aldelyde- 3- phosphate- dehydrogenase,GAPDH)作内参照。结果 :正常 SD大鼠心脏各部位及血管 (动脉、静脉 ) ECEm RNA的相对含量 (ECE/ GAPDH)分别为 :左房 1.17± 0 .2 1,右房 1.144± 0 .144 ,左室 0 .96 6± 0 .0 91,右室 0 .95 6±0 .0 95 ,室间隔 1.0 8± 0 .138,动脉 0 .742± 0 .174,静脉 1.5 38± 0 .2 37,静脉含量最高 ,动脉含量最低 ,而心脏各部位心房比心室含量高。结论 :正常 SD大鼠心脏各部位及血管均有 ECE m RNA的表达 ,与左室比较 ,静脉 ECE m RNA的表达 (P<0 .0 1) ,左房右房亦较高 ,动脉含量低 (P<0 .0 5 ) ,而左室、右室、室间隔间无量著差异 ,由高到低的趋势为 :静脉>左房≈右房 >室间隔≈左室≈右室 >动脉。 Objective:To investigate the distribution of endothelium-converting enzyme(ECE) mRNA in cardiovascular system of normal SD rats.Medthods:With glyceraldehyde-3-phosphate-dehydrogenase(GAPDH) as a internal standard,ECE mRNA in rat different myocardiac tissues and vascular (artery,vein) were determined by semi-quantitive RT-PCR.Results:The ECE mRNA expression of normal SD rats in cardiovascular from vein(V),left atrium(LA),right atrium(RA),left ventricle(LV),right ventricle (RV) intraventricular septum(IVS) and artery(A) were 1.538±0.237,1.17±0.21,1.144±0.144,0.966±0.091,0.956±0.095,1.08±0.138,and 0.742±0.174 respectively.ECE mRNA expression was the highest in vein,the lowest in artery.ECE mRNA expression of atrium is higher than that of ventricle in heart.Conclusion:ECE mRNA expressed in all parts of adult rats cardiovascular.By comparing with left ventricle,the expression of ECE mRNA in vein is the highest(P<0.01),atrium is higher,artery is the lowest(P<0.05),and there are no differences in left ventricle,right ventricle,intraventricular septum.The rank from the highest to the lowest was V>LAIVSLVRV>A.
