摘要
目的 研究神经生长因子 (NGF )在烧伤创面愈合中的作用 ,探索烧伤创面愈合机制。方法2 0 kg小白家猪 6只为实验对象。用控温控压电烫仪在每只猪背部制成 2 4个直径为 2 .5 cm的圆形深 度烧伤创面。分为四组 ,分别为创面局部应用 1、2 .5和 5μg/m l的 NGF实验组和不含 NGF的生理盐水对照组。在用药后 3、5和 9天分别取创面组织进行表皮生长因子 (EGF)受体 ,EGF,NGF受体 ,NGF,CD6 8,CD3免疫组织化学检测。结果 六项免疫组织化学检测结果 ,在各实验组均见有不同程度的表达 ,且第 5和第 9天两时间点较为明显。结论烧伤创面局部应用 NGF后 ,外源性 NGF直接作用于创面组织 ,调控释放内源性 NGF及其它生长因子 ,生长因子受体的释放增多 ,加速创面的愈合。
Objective To investigate the effect of nerve growth factor(NGF) on the burn wound healing and to study the mechanism of burn wound healing. Methods Six domestic pigs weighting around 20 kg were used as experimental animals. Twenty-four burn wound, each 2.5 cm in diameter, were induced on every pigs by scalding. Three different concentrations of NGF,1 μg/ml, 2.5 μg/ml, 5 μg/ml were topically applied after thermal injury,and saline solution used as control group. Biopsy specimens were taken at 3,5 and 9 days following treatment and immunohistochemistry method was used to detect the epidermal growth factor(EGF), EGF receptor(EGF-R), NGF, NGF receptor(NGF-R),NGF, NGF-R,CD 68 and CD 3. Results The experssion of EGF, EGF-R, NGF,NGF-R CD 68 and CD 3 were observed in the experimental group, especially at 5 and 9 days, no expression of those six items in the control group. Conclusion NGF can not only act directly on burn wound, but also modulate other growth factors on the burn wound to accelerate the healing of burn wound.
