摘要
采用HPLC法测定大鼠血浆中曲安奈德(TA)含量,通过对线性关系、精密度、回收率、重复性及稳定性进行试验分析,找出快速有效的含量测定方法.结果表明,采用Dionex C18柱,流动相为甲醇-水(57∶43),流速为1.0mL/min,检测波长为240nm,柱温为35℃,以曲安西龙为内标进行测定,在0.1~64.0μg/mL浓度范围内线性关系良好,日内、日间精密度RSD均低于3%,方法回收率均在90%以上.该法可用于大鼠血浆中曲安奈德含量的测定.
To develop a HPLC method for the determination of triamcinolone acetonide(TA) in rats plasma,the linear relationship,the intra-and inter-day RSD,the method recovery,the reproducibility and the stability were respectively analyzed.The results showed that the method had a good linear relationship between 0.1 and 64.0 μg/mL,the intra and inter-day RSD were less than 3.0%,and the method recovery was more than 90%,while conditions were as follows: a Dionex C18 column was used;the mobile phase,methanol and water(57∶43,v/v);the flow rate,1.0 mL/min;the UV detection wavelength at 240 nm;the column temperature at 35 ℃;the internal standard substance,triamcinolone tablets.The method is reliable and rapid for the analysis of the concentration of TA in rats plasma.
