倍他福林对大鼠离体子宫平滑肌收缩作用的影响研究 被引量:1

摘要 目的:研究倍他福林对大鼠离体子宫平滑肌收缩作用的影响。方法:制备未孕和妊娠大鼠离体子宫平滑肌标本,分别设为未孕空白(NPC)组、妊娠空白(PC)组、倍他福林未孕(BNP)组、倍他福林妊娠(BP)组、利托君未孕(RNP)组和利托君妊娠(RP)组(n=6)。采用累积加药的方法,每隔15min加药1次,BNP组和BP组分别加入倍他福林至10-10、10-9、10-8、10-7、10-6、10-5mol/L,RNP组和RP组分别加入利托君至10-9、10-8、10-7、10-6、10-5、3×10-5mol/L,NPC组和PC组不加任何药物。记录各组平滑肌105min内的收缩幅度,计算收缩率、半数抑制浓度(IC50)的-lg值(PIC50)和最大抑制效应(Emax)。结果:与NPC组和PC组比较,BNP组和BP组对应模型平滑肌累积加入倍他福林10-10~10-5mol/L后的收缩率明显降低(P<0.05),且浓度间差异有统计学意义(P<0.05);倍他福林对BNP组和BP组平滑肌收缩作用的PIC50分别为7.74±0.12和7.92±0.07,Emax分别为(80.32±5.49)%和100%。与NPC组和PC组比较,RNP组和RP组对应模型平滑肌累积加入利托君10-9~3×10-5mol/L后的收缩率明显降低(P<0.05),且浓度间差异有统计学意义(P<0.05);利托君对RNP组和RP组平滑肌收缩作用的PIC50分别为7.28±0.16和6.98±0.13,Emax分别为(63.63±6.28)%和(65.48±6.93)%。结论:倍他福林和利托君均能浓度依赖性抑制未孕和妊娠大鼠离体子宫平滑肌的自发性收缩,倍他福林的抑制作用强于利托君。 OBJECTIVE: To investigate the effects of betaphrine on smooth muscle contraction of rat isolated uterus. METH- ODS: The isolated uterine smooth muscles samples of non-pregnant and pregnant rats were prepared. They were divided into non-pregnant blank (NPC) group, pregnant blank (PC) group, betaphrine non-pregnant (BNP) group, betaphrine pregnant (BP) group, ritodrine non-pregnant (RNP) group and ritodrine pregnant (RP) group (n=6). The concentration of medicine was accumu- lated every 15 min in those groups; by adding betaphrine, the concentration of medicine was accumulated to 0^-10、10^-9、10^-8、10^7、10^6、and 10^-5 mol/L in BNP group and BP group, respectively; by adding ritodrine, the concentration of medicine accumulated to 0^-10、10^-9、10^-8、10^7、10^6、10^5 and 3×10^-8 mol/L in RNP group and RP group, respectively; no medicine was given to NPC and PC group. The range of myometrial contraction was recorded within 105 rain. Myometrial contraction rate, -lgIC50 (PIC50) and Emax were measured. RESULTS : Compared with NPC group and PC group, the contraction rate of smooth muscles deceased significantly in BNP group and BP group after 10^-10-10^-9 mol/L betaphrine was added in smooth muscles of corresponding model (P〈0.05), there was significant difference among different concentrations of betaphrine (P〈0.05) ; PIC50 of betaphrine in BNP group and BP group were 7.74 ± 0.12 and 7.92 ± 0.07, and Emax of betaphrine were (80.32 ± 5.49)% and 100%, respectively. Compared with NPC group and PC group, the contraction rate of smooth muscles deceased significantly in RNP group and RP after 10^-9-3×10^-5 mol/L ritodrine was added in smooth muscles of corresponding model (P〈0.05), there was significant difference among different concentrations of ritodrine (P〈0.05) ; PIC of ritodrine in RNP group RP group were 7.28 ± 0.16 and 6.98 ± 0.13, and Emax of rito- drine were (63.63 ± 6.28) % and (65.48 ± 6.93 ) %, respectively. CONCLUSIONS : Betaphrine and ritodrine can inhibit spontaneous contraction of isolated uterine smooth muscle in non-pregnant and pregnant rats, in concentration-dependent manner, and inhibitory effect of betaphrine is better than that of ritodrine.
