摘要
用MCM-41介孔分子筛固定α-淀粉酶、葡萄糖淀粉酶和葡萄糖氧化酶,将其用于流动注射化学发光分析体系,可实现对淀粉的快速检测。在酶柱使用温度为55℃,淀粉溶液的酸度为pH=5.0,淀粉溶液的流速为2 r/min的条件下,体系发光强度与淀粉浓度呈良好线性关系,测定的线性范围为1~150mg/L,相关系数0.999 8,检出限为0.3 mg/L,回收率为97.2%~102.2%,连续10次测定的RSD为2.1%,单次测定能在1 min内完成。方法简便、快速、灵敏、准确,对8个样品的测定结果与国标方法的测定结果无显著性差异。
In this paper, a- amylase, glucoamylase and glucose oxidase were immobilized by MCM -41 meso- porous molecular sieves. With its application to flowing injection chemiluminescence system, starch could be rapidly determined online. Used in the enzyme column temperature of 55 ℃, the starch solution acidity of pH = 5.0, the flow rate of the starch solution under conditions of 2 r/min, the starch concentration and system luminous intensity showed a good linear relationship ,the liner range of its concentration is 1 - 150 mg/L with the correlation coefficient of 0. 999 8, and detection limit is 0.3 mg/L. The recoveries are 97.2%- 102.2% and the relative standard deriation is 2.1% with 10 continuous assays. A complete analysis could be performed in 1 rain. The determination results of 8 simples show no significant difference between this method and the standard method, while it is simple, rapid, sensitive and accurate.
