卵巢上皮性癌中表达Tie2单核巨噬细胞的研究 被引量:2

Study on Tie2-expressing monocytes /macrophages in patients with epithelial ovarian cancer

摘要 目的 探讨卵巢上皮性癌(EOC,epithelial ovarian cancer)患者外周血和癌旁腹膜中表达Tie2单核巨噬细胞(Tie2-expressing moncytes/macrophages,TEMs)的分布情况。方法 2011年7月至2012年12月于同济大学附属第一妇婴保健院,分离正常成年女性、卵巢良性肿瘤及EOC患者外周血单核细胞;RT-PCR检测Tie2 mRNA的表达情况,流式细胞术检测Tie2^+细胞占CD14^+外周血单个核细胞的比例;免疫荧光技术分析TEMs在良性卵巢肿瘤、EOC原位灶及癌旁腹膜中的浸润情况。结果 正常成年女性、良性卵巢肿瘤及EOC患者外周血单核细胞在mRNA和蛋白水平均表达Tie2;良性卵巢肿瘤及EOC患者外周血TEMs的比例分别为(9.41±5.93)%、(1.82%~23.45%)和(7.99±3.90)%(1.71%~15.03%),较正常对照组(4.13±2.86)%(1.18%~9.56%)比例增加,差异有统计学意义(P<0.05);形态学研究证实EOC患者癌旁腹膜中浸润着数量丰富的TEMs(15.17±8.84)%(2.27%~25.81%),而良性卵巢肿瘤患者腹膜中几乎没有TEMs的浸润。结论 EOC患者外周血、原位灶及癌旁腹膜组织中存在表达Tie2的单核巨噬细胞;TEMs比例在EOC患者外周血、腹膜局部显著上升,有利于肿瘤微环境中血管的生成,从而参与调控卵巢癌的进展。 Objective To investigate the frequency of Tie2-expressing monocytes (TEMs) in the peripheral blood and peritoneum of epithelial ovarian cancer. Methods Between Jul. 2011 and Dec. 2012, monoeytes in peripheral blood were isolated from female healthy donors , patients with benign ovarian tumor and patients with EOC. The mRNA expression of Tie2 was measured by RT-PCR. Flow cytometry was used to detect the rate of TEMs in CD14 + monocytes in peripheral blood. The infiltration of TEMs in peritoneum were detected by immunofluoreseence. Results The mRNA and protein of Tie2 expression were identified in monocytes isolated from female healthy donors, patients with benign ovarian tumors and EOC. The frequency of TEMs ranged from 1.82% to 23.45% (9.41 ± 5.93 ) % in benign ovarian tumors and 1.71% to 15.03% (7.99 ± 3.90) % of CD14 + PBMC ( peripheral blood monocytes) in EOC patients. Compared with 1.18%- 9. 56% (4. 13 ±2. 86)% in healthy donors , the frequency of TEMs was significantly higher in patients with benign ovari- an tumors and EOC ( All P 〈 0. 05 ). It was observed that the infiltration of TEMs [ ( 15.17 ± 8.84) % (2. 27%- 25.81% ) ] in the peritoneum of EOC patients. In the mean time, no TEMs was shown in peritoneum in benign ovarian tumors. Conclusion TEMs could be detected in the peripheral blood and peritoneal of EOC patients. The frequency of TEMs was significantly higher in peripheral blood and peritoneal of EOC patients.
