摘要
目的 :探讨内毒素 (L PS)对体外培养鼠微血管内皮细胞 (MEC)的影响。方法 :应用体外培养的鼠肺MEC,观察 L PS对其分泌功能及 s ICAM 1和 s E sel表达的影响。结果 :1在 L PS作用下 ,ET 1和 NO6 0分钟高于对照组 ,180分钟达高峰 ,高水平状态持续到 2 40分钟 ,36 0分钟有下降趋势 ,L PS高峰浓度为1mg/L。2 s ICAM 1在 L PS作用后 6 0分钟表达迅速上调 ,呈时间、剂量依赖性 ,L PS高峰浓度为 0 .0 1m g/L。3s E sel在 L PS作用后 180分钟表达才明显上调 ,而 L PS高峰浓度仅为 0 .0 0 1mg/L。结论 :L PS能引起体外培养的鼠肺 MEC分泌 ET 1和 NO增加 ,并有一定的时间、剂量依赖性 ;L PS可活化体外培养 MEC导致MEC表面 s ICAM 1和 s E sel过度表达 ,并能间接反映 MEC受损情况 ,将 s ICAM 1和 s E sel两者结合分析 ,能较全面地反映受损情况。
Objective:To investigate the effects of lipopolysaccharide (endotoxin,LPS) on microvascular endothelial cells (MEC) in vitro.Methods:Rat MEC was used to observe the excretion and expression of soluble intercellular adhesion molecule1 (sICAM1) and soluble Eselectin (sEsel) in vitro.Results:①Following LPS stimulation (peak concentration in 1 mg/L),endothelin1(ET1) and nitric oxide(NO) levels significantly elevated at 60 minutes compared to controls,peaking at 180 minutes and decreasing at 360 minutes. ②The sICAM1 expression enhanced quickly at 60 minutes after LPS treatment,showing doseand timedependent response (peak LPS concentration in 0 01 mg/L).③The sEsel expression was observed only at 180 minutes after LPS treatment and the peak concentration of LPS was 0 001 mg/L.Conclusions:LPS stimulation may lead to increased excretion of ET1 and NO in MEC of rats,showing doseand timedependent response.Also,LPS can activate pulmonary MEC in culture,thereby resulting in overexpression of sICAM1 and sEsel on the surface of MEC.In addition,expression of sICAM1 and sEsel on surface of MEC may serve as useful parameters to indirectly reflect MEC injury.
