摘要
目的 :研究烧伤早期心肌抑制性 G蛋白 α亚基 (Giα)含量的变化 ,探讨其在烧伤后心肌舒缩功能下降中的作用。方法 :采用 30 %体表面积 度烫伤大鼠模型 ,测定伤前及伤后左心室收缩压 (L VSP) ,左心室压力最大上升与下降速率 (L V± dp/dtm ax)。采用 Western杂交法和放射免疫法分别测定大鼠心肌 Giα及腺苷酸环化酶 (AC)的活性。结果 :与对照组比较 ,烫伤后心功能指标显著降低 ,烫伤后 12、2 4和 48小时心肌 Giα分别升高6 9.5 % (t=5 .79,P<0 .0 1)、37.8% (t=3.89,P<0 .0 1)和 2 9.1% (t=3.42 ,P<0 .0 5 ) ,AC活性降低。结论 :烫伤后 Giα含量升高是导致腺苷酸环化酶信号转导系统功能障碍及心肌舒缩功能下降的重要原因之一。
Objective:To investigate the alteration of Gi proteins and its potential role in mediated cardiac contractile dysfunction after severe thermal injury in rats.Methods:Contraction relaxation properties of left ventricle (LV) were studied in rats subjected to a 30% total body surface area fullthickness thermal injury.Levels of Giα protein in sarcolemma were measured by Western blot analysis,and adenyl cyclase activities were determined by radioimmunoassay.Results:Cardiac function parameter significantly decreased following thermal injury.Compared with the control group,Giα levels in sarcolemma at 12,24,and 48 hours postburn increased by 69 5% (t=5 79,P<0 01),37 8%(t=3 89,P<0 01) and 29 1%(t=3 42,P<0 05) respectively.Activities of adenyl cyclase decreased after injury.Conclusions:The increased expression of cardiac Gi protein may,at least in part,contribute to the dysfunction of adenyl cyclase signal transduction system and the impaired cardiac function after severe burns.
