环磷酰胺免疫抑制小鼠模型的建立及其在肾包膜下移植中应用 被引量:8

摘要 :目的 :建立一种适用于肿瘤药敏人癌肾包膜下移植法 (SRCA)的免疫抑制小鼠模型 ,克服普通小鼠的免疫排异反应及裸鼠费用昂贵的缺陷。方法 :环磷酰胺处理 BAL B/ C小鼠 ,建立环磷酰胺免疫抑制小鼠模型。以裸鼠为标准对照组 ,环磷酰胺小鼠和 BAL B/ C小鼠为实验组 ,用同一新鲜人癌标本进行肾包膜下移植。比较移植后 4d、6 d、8d、1 0 d各组间移植物未初体积差 (△ TS)、移植癌存活率和病理分级。结果 :环磷酰胺免疫抑制小鼠与裸鼠移植癌 4d、6 d、8d均表现出持续正性生长 ,两组△ TS,移植瘤存活率 ,病理分级分析比较均无显著性差异 (P>0 .2 0 )。而分别与正常免疫力小鼠比较 ,差别有显著性 (P<0 .0 5 )。结论 :环磷酰胺免疫抑制小鼠表现为有效的免疫抑制特性 ,其人癌移植物8d内持续良好生长 。 Objective:To eliminate the rejective reaction of host-versus-graft in the SRCA,a new kind of immunosuppressed mice model was established.Method:BALB/c mice was pretreated with CTX to establish the immunosuppressed mice model.SRCA was carried out simultaneously with fresh explants of human carcinoma in three groups-the CTX-immunosuppressd mice athgmic nude mice and immuno-persistence of viable tumor cell and the patho-histology classification were observed at day 4,5,6,10 after SRCA.Result:The xenograft grew constantly within 8 days after SRCA in CTX mice and nude mice,the mean of △TS,the persistentence of viable tumor cell and the histology classification is similar each other(P>0.20).But the all three indexes above were different from that of immuno-competent mice significantly(P>0.05).Conclusion:CTX can inhibit the rejection of host-versus-graft effectively.Xenografts under renal capsule of CTX immunosuppressed mice could grew constantly in the same way as in nude mice within 8 days.It was more suitable for lab and clinical research than nude mice because of it′s lower cost and easier to keep.
