高效毛细管电泳法分析茯苓多糖中的单糖组成 被引量:20

摘要 本文旨在建立简单、快速分析茯苓多糖中单糖组成的高效毛细管电泳法。将茯苓多糖水解,经PMP衍生化,采用高效毛细管电泳分析。电泳条件:缓冲体系为80mmol/L硼砂溶液(pH9.5);毛细管柱(25μm×42cm),分离电压18kV,检测波长254nm,进样高度10cm,进样时间25s。结果显示,茯苓多糖中含葡萄糖、甘露糖和半乳糖的物质的量之比为2.657∶1.000∶2.362。该方法具有简单、快速、准确、重现性好,可用于测定茯苓多糖中单糖的组成。 To develop a simple and rapid high performance capillary electrophoresis (HPCE) for the determination of monosaeeharide composition in Poria cocos polysaceharides. Polysaceharide was hydrolyzed, then 1-phynel-3-methyl-5- pyrazolone(PMP) was used as derivatization reagent and the released monosaccharide derivatives were analyzed by HPCE. The optimized conditions were achieved with 80 mmol/L Borax (pH 9.5), capillary electrophoresis was performed using a 25 ~m^42 cm fused silica capillary column. Separation voltage was 18 kV, detection wavelength was set at 254 nm, and the injection time was 25 s at a height of 10 cm. The results showed that Poria cocos polysaccharide was composed of Glu:Man:Gal=2.657:l.000:2.362. The method can be used for the quality control of the Poria cocos polysaccharides with good precision.
