摘要
目的 :探讨基因表达谱芯片技术在高通量筛查肿瘤相关基因群及研究胰腺癌分子病理学变化中的应用价值。方法 :应用含有 40 96条人类全长基因的 c DNA表达谱芯片 ,对 3例临床切除的胰腺癌及正常胰腺标本的基因表达谱进行分析。 结果 :在 3例胰腺癌和正常胰腺组织中均有差异表达的基因 398条 ,其中新基因 2 89条 ,老基因 10 9条。从老基因中筛选出有显著表达差异基因 37条 ,其中在胰腺癌组织中上调基因 2 0条 ,下调基因 17条。 结论 :运用本基因表达谱芯片对基因表达谱进行分析 ,能够有效筛查出新的胰腺癌相关基因。
Objective: To evaluate the role of expression microarray analysis in molecular pathophysiology of pancreatic cancer, and in the identification of novel cancer associated genes. Methods: A series of expression microarray analysis of human pancreatic cancer were initiated by cDNA microarray which representing a set of 4 096 human genes. Results: By applying this cDNA microarray we identified 398 differentially expressed genes in all 3 clinical pancreatic cancer samples, and we further identified 20 upregulated and 17 downregulated genes for functional analysis. Conclusion: cDNA microarray for analysis of gene expression patterns is a powerful method to identify novel cancer associated genes, and it is helpful for rapid exploring of their role in clinical pancreatic cancer.
