Bcl-xl和Bcl-2基因在人和兔心肌细胞中的同源性研究

摘要 目的 探讨兔和人的有核细胞中Bcl-xl和Bcl- 2基因的同源性。方法 用人的Bcl-xl和Bcl- 2基因序列来扩增兔心肌细胞的DNA ,纯化其扩增产物 ,然后进行测序 ,再测定人与兔心肌细胞内Bcl-xl和Bcl- 2基因的同源性。结果 兔心肌细胞内Bcl-xl基因的扩增产物长度为 16 2个碱基 ,与人类Bcl-xl基因的碱基序列进行比较 ,在扩增的 15 9个碱基片段中有 15 1个碱基相同 ,其同源性为 95 0 % ;而兔心肌细胞内Bcl- 2基因的扩增产物长度为 2 88个碱基 ,与人类Bcl- 2基因mR NA的碱基序列进行比较 ,在扩增的 199个碱基片段中有 176个碱基相同 ,其同源性为 88 4%。结论 兔和人的有核细胞中Bcl-xl和Bcl- 2基因有很好的同源性 ,在研究兔心肌细胞这两种基因的改变时 ,完全可以用人的Bcl-xl和Bcl- 2基因序列(引物 )来扩增 。 Objective To investigate Bcl-xl and Bcl-2 gene homogeneity in rabbit cardiac myocytes and human karyotes.Methods DNA in rabbit cardiac myocytes were amplificated by using human Bcl-xl gene and Bcl-2 gene sequence.The amplificated products of DNA in rabbit myocytes were purificated before sequenced.The homogenous of Bcl-xl gene and Bcl-2 gene in rabbit myocardial cells and human karyotes were analysed.Results The length of the amplificated products of Bcl-xl gene in rabbit myocardial cells was 162 bases.The 95.0% of Bcl-xl gene in rabbit myocytes was homogeneous with human Bcl-xl gene in 159 bases sequences.And the length of the amplificated products of Bcl-2 gene in rabbit myocardial cells was 288 bases.The 88.4% of Bcl-2 gene in rabbit myocytes was homogenous with human Bcl-2 gene in 199 bases.Conclusions There were good homogeneities of Bcl-xl gene and Bcl-2 gene in between rabbit myocytes and human karyotes.Human Bcl-xl and Bcl-2 gene sequence,therefore,is completely used to amplificate both genes in rabbit cardiac myocytes when studying,which offerefore,basic theory for further study. [
