摘要
目的 采用一种新的多聚阳离子化合物——多聚乙酰亚胺 (polyethylenim ine,PEI) ,作为将外源性基因转入骨骼肌细胞的介导物。 方法 骨骼肌细胞取自新生 SD大鼠 ,经胰酶消化后培养 ,将 L ac Z基因 DNA - PEI复合物导入骨骼肌细胞内 ,最后用 X- gal组织化学染色检测已转染的细胞。 结果 PEI可作为介导剂使 L ac Z基因在培养的骨骼肌细胞中表达 ,其表达率可达到 30 %。 结论 PEI在真核细胞中实施基因转移是一种有效方法 ,为基因治疗。
Objective\ By using the polycation polyethylenimine(PEI) to explore the possible new agents for foreign gene transfected into muscle cells in vitro. Methods\ Muscle cells were isolated from SD rat and trypsinized for primary culture. The LacZ gene DNA\|PEI complex was transfected into muscle cells. LacZ(a reporter gene) expression was detected by X\|gal immunohistochemical stain. Results\ PEI was demonstrated to be a useful reagent for gene delivery and LacZ expression efficiency was about 30%. Conclusions\ PEI\|mediated transfer is a simple, efficient method which could be a new approach for gene therapeutics.\;
