摘要
目的:探讨右归丸含药血清体外诱导大鼠骨髓基质干细胞分化为神经元样细胞的最佳浓度。方法:用DMEM/F-12培养液培养Wistar大鼠骨髓基质干细胞(BMSCs),采用不同浓度右归丸含药血清对其进行体外诱导,并用MTT法检测各组细胞生长情况,用免疫细胞荧光方法计算各组神经元样细胞阳性数加以比较。结果:BMSCs在2.5%含药血清培养液中活力最好。结论:2.5%右归丸含药血清培养液是诱导BMSCs分化实验的最佳浓度。
Objective : To establish the optimal concentration of the serum containning Yougui Pill which could induce rat bone marrow stromal stem cells differentiated into neurons - like cells in vitro. Methods : Rat bone marrow stromal stem cells (BMSCs) were cultured in DMEM/F - 12 medium and inducted into neurons - like cells by serum with a different concentration of Yougui Pill,then the proliferation ability of each group cells was investigated by MTT detection. Immunohistochemical staining and image analysis were used to detect the number of the positive neurons. Results:BMSCs reached best activity in serum eontainning 2.5 % Yougui Pill. Conclusion:BMSCs could be inducted into neurons-like cells in vitro by serum containning Yougui Pill with 2.5% as the best concentration.
