摘要
目的:研究西洋参茎叶总皂苷(PQS)对心肌梗死后心力衰竭大鼠血流动力学及血浆脑钠肽(BNP)的影响。方法:健康雄性SD大鼠70只,随机抽取假手术(sham)组10只,结扎余60只大鼠左冠状动脉前降支复制心肌梗死模型。术后24 h,成活的心肌梗死大鼠随机分为急性心肌梗死(AMI)组及PQS[50 mg/(kg·d)、100 mg/(kg·d)、200 mg/(kg·d)]组各10只。各用药组用饮用水10 mL/(kg·d)溶解药物灌胃4周,sham组及AMI组予等量饮用水灌胃4周。术后4周,颈动脉插管检测血流动力学,应用BIOSITE干式快速心力衰竭/心肌梗死诊断仪测定血浆BNP含量。结果:各实验组之间心率无显著性差异(P>0.05)。与sham组比较,AMI组平均颈动脉压、左室收缩压、左室收缩压最大上升速率及左室收缩压最大下降速率绝对值均显著降低,左室舒张末压及血浆BNP水平显著升高;PQS低、中、高剂量组心肌梗死后大鼠功能障碍均明显改善,与AMI组比较,平均颈动脉压、左室收缩压、左室收缩压最大上升速率及左室收缩压最大下降速率绝对值均显著升高,左室舒张末压及血浆BNP水平显著降低(P<0.05)。结论:PQS剂量依赖性减轻大鼠心肌梗死后心功能障碍,具有抗心力衰竭的作用。
Objective:To investigate the effect of Panax quinquefolium saponin (PQS)on hemodynamics and plasma brain natri- uretic peptide level. Methods:lO rats were randomly divided from 70 healthy male rats as the sham group, and the remaining 60 rats were subjected to permanent left coronary artery ligation. 24h after ligation, surviving rats were divided randomly to the AMI group, the PQS 50 mg/( kg · d), the PQS 100 mg/( kg · d)and the 200 mg/( kg · d)groups (n = 10 ). Rats in drug treatment groups were gavaged with PQS powder dissolved in water[ 10 mL/(kg · d) ] ,and rats of the sham group and the AMI group re-ceived an equivalent volume of vehicle (water) daily. After 4 weeks, the hemodynamic paraments were measured by cannulating through carotid artery;the plasma brain natriuretic peptide level was examined by BIOSITE dry fast heart failure or myocardial in-farction diagnosis instrument. Results : There was no significant difference in heart rate among each group ( P 〉 0. 05 ). Compared with the sham rats, the mean arterial pressure,left ventricular systolic pressure, the maximal rates of rise and decline of left ven-tricular pressure of the rats from AMI group were all significantly lower( P 〈 0. 05 ), however, the left ventricnlar end-diastolic pressure and plasma BNP levels were significantly elevated (P 〈 0. 05 ). Compared with the AMI rats, the mean arterial pressure, left ventricular systolic pressure, the maximal rates of rise and decline of left ventricular pressure of the rats from PQS treated groups were all significantly increased( P 〈 0. 05 ) and the left ventricular end - diastolic pressure and plasma BNP levels were sig- nificantly decreased (P 〈 0. 05 ). Conclusion:PQS dose - dependently reduced heart dysfunction in the heart failure rats after myo-cardial infarction.
