薯蓣皂苷元抗骨肉瘤机制及与Wnt/β-catenin信号通路的关系 被引量:4

摘要 目的探讨薯蓣皂苷元抗骨肉瘤机制及与Wnt/β-catenin信号通路的关系。方法通过动物实验检测薯蓣皂苷元对移植人骨肉瘤荷瘤鼠肿瘤增重的影响,用RT-PCR法、Envision法研究薯蓣皂苷元对人骨肉瘤U-2OS细胞及荷瘤裸鼠β-catenin的影响。结果 400、200、100 mg/kg薯蓣皂苷元灌胃对荷瘤裸鼠肿瘤生长有明显的抑制作用,呈量效关系,抑瘤率30%~60%;100~400 mg/kg呈剂量依赖性抑制U-2OS细胞β-catenin的表达;400 mg/kg抑制荷瘤裸鼠β-catenin的表达。结论薯蓣皂苷元抗骨肉瘤机制与抑制Wnt/β-catenin信号通路有关。 Objective To investigate the relationship between the anti-osteosarcoma mechanism of diosgenin and Wnt/β-catenin signal pathway. Methods The effect of diosgenin on tumor growth of nude mice bearing human osteosarcoma xenograft in vivo was examined with animal experiments, and the effects of diosgenin on human osteosarcoma U-20S cells and β-eatenin of nude mice were analyzed by RT-PCR and Envision method. Results Diosgenin with dose of 400, 200 and 100 mg.kg-1 had significant inhibitory ef- fect on tumor growth of nude mice, this inhibitory effect showed a dose-effect relationship, and the inhibition rates were 30%-60%. 100-400 mg.kg-1 diosgenin inhibited the expression of β-catenin in U-20S cell line in a dose-dependent manner. 400 mg .kg-1 diosgenin inhibited the expression of β-catenin in nude mice bearing human osteosarcoma xenograft. Conclusion The anti-osteosarcoma mechanism of diosgenin may be related with the effect of inhibiting Wnt/β-catenin signal pathway.
