摘要
目的观察针刺后海穴对自闭症模型大鼠大脑皮层M1区和海马CA1区PSD-95蛋白表达的影响。方法采用孕鼠腹腔注射丙戊酸钠(VPA)的方法建立自闭症大鼠模型,分为空白组、模型组、非穴组、针刺组,每组6只。针刺组取后海穴,用0.5寸一次性针灸针直刺0.3寸,行平补平泻提插手法1 min后出针,10天为1个疗程,共治疗3个疗程;非穴组选取大鼠右侧胁下固定非经非穴点,余操作同针刺组;空白组、模型组不予任何干预;应用免疫组化技术观察4组大鼠大脑皮层M1区和海马CA1区PSD-95蛋白的表达。结果大鼠大脑皮层M1区:与空白组比较,模型组PSD-95蛋白表达有显著性差异(P<0.05);与模型组比较,针刺组PSD-95蛋白表达有显著性差异(P<0.05)。海马CA1区:与空白组比较,模型组PSD-95蛋白表达有显著性差异(P<0.05);与模型组比较,针刺组PSD-95蛋白表达有显著性差异(P<0.05)。结论针刺后海穴可提高自闭症模型大鼠大脑皮层M1区和海马CA1区PSD-95蛋白的表达。
Objective To observe the effect of acupuncturing at Houhai on expression of PSD-95 protein in cerebral cortex M1 area and hippocampus CA1 area of autism model rats. Methods The pregnant rats were injected intraperitoneally with sodium valproate (VPA) to establish rat models of autism, then rats were randomly divided into blank group, model group, non-acupoint group and acupuncture group, and each group had 6 rats. The acupuncture group was treated with acupuncturing at Houhai (the depth for perpendicu- lar needling of the point was 0.3 cun), then received lifting and thrusting techniques for 1 minute and with- draw the needle, 10 days as a course of treatment, and the group received 3 courses of treatment totally. The non-acupoint group received acupuncturing at the fixed non-point under rat fight flank. The blank group and model group did not received any intervention. The expression of PSD-95 protein in cerebral cortex M1 area and hippocampus CA1 area of 4 groups were observed by immunohistochemical technique. Results There was a significant difference in expression of PSD-95 protein in cerebral cortex M1 area between blank group and model group (P〈0.05). There was a significantly difference in expression of PSD-95 protein in cerebral cortex M1 area between model group and acupuncture group (P〈0.05). There was a significant difference in expression of PSD-95 protein in hippocampus CA1 area between blank group and model group (P〈0.05). There was a significant difference in expression of PSD-95 protein in hippocampus CA1 area between model group and acupuncture group (P〈0.05). Conclusion Acupuncturing at Houhai can increase the expres- sion of PSD-95 protein in cerebral cortex M1 area and hippocampus CA1 area of autism model rats.
