沙葱多糖的提取纯化及纯度鉴定 被引量:3

摘要 采用热水浸提法提取沙葱多糖,三氯乙酸法除蛋白,活性炭脱色,Sephadex凝胶柱层析纯化,收集单一峰溶液命名为SCSP,对纯化产物SCSP分别采用紫外分光光度法、纸层析法、比旋光度法、凝胶柱层析4种方法联合检验其纯度,结果表明,在波长260nm和280nm处无核酸和蛋白的明显吸收,纸层析见单一染色海蓝色斑点,比旋光度测定结果均一致为+51.36°,凝胶柱层析结果显示出现了单一狭窄对称峰,以上纯化结果综合表明SCSP为均一组分多糖。 The Hot--water extraction was used to extract Allium mongolicum Regel polysaccharide, the Sevage method was used to remove the protein from Allium Mongolicum Regel Polysaccharide, thus active carbon decoloring process was optimized. Decolored Allium mongolicum Regel polysaccharide was purified with SephadexG-100 column chromatography, Collected singular peak liquor named SCSP. Its purity was examined by means of UV spectra, paper chromatography, specific rotation and gel column chromatography. The result shows that SCSP have not obvious absorped to nucleic acid and protein at 260nm and 280nm of wave length, paper chromatography displayed singular colored spot, the similar de- termined results of specific rotation were + 51. 36°, and gel column chromatography displayed singular peak. Above results comprehensive shows that SCSP is a homogeneous neutral polysaccharide.
