激光针灸对CIA大鼠滑膜组织病理改变的影响研究 被引量:4

摘要 目的研究激光针灸对Ⅱ型胶原蛋白诱导的关节炎(CIA)模型大鼠滑膜组织病理改变的影响。方法将大鼠随机分为激光针灸50、100、150mW组,并设正常组、模型组和药物组作对照。激光光斑直径0.3cm,每次照射10min,穴位选择足三里和肾俞,干预10d后取材,制作踝关节病理切片,HE染色,光镜下观察滑膜组织炎性细胞浸润、纤维组织增生、滑膜细胞增生及巨噬细胞增生情况。结果模型组在滑膜细胞增生(P〈0.001)、炎性细胞浸润(P〈0.01)、巨噬细胞增生(P〈0.01)、纤维组织增生(P〈0.001)的分级评分和总积分(P〈0.001)方面均较正常组显著降低,差异具有统计学意义。治疗后,与模型组相比,各治疗组在以上评分方面均显示出不同程度的降低,差异具有统计学意义。其中,激光针灸50mW组的纤维组织增生评分(P〈0.01),激光针灸100mW组的滑膜细胞增生评分、纤维组织增生评分和总积分(均P〈0.01),激光针灸150mW组的滑膜细胞增生评分(P〈0.05)、纤维组织增生评分(P〈0.01)和总积分(P〈0.05)。治疗组评分均显著优于模型组;而且激光针灸100mW组的滑膜细胞增生评分显著优于药物组,差异具有统计学意义(P〈0.05)。结论激光针灸对CIA大鼠滑膜组织的滑膜细胞增生、纤维组织增生等病理改变有明显抑制作用,其作用效果与激光的参数设置有关。 Objective To study the influence of laser acupuncture on the pathological change of synovial tissue in the Type Ⅱ collagen-induced arthritis (CIA) model rat. Methods The rats were randomly divided into normal group, model group, drug group, 50mW laser acupuncture group, 100mW laser acupuncture group and 150 mW laser acupuncture group, with the laser spot diameter of 0.3cm. Rats were exposed to the laser for 10 minutes each time, and Zusanli and Shenshu were chosen as the test acupoints, Material was drawn10 days after the intervention, and pathological section of ankle joint was made and stained with HE. The inflammatory cell infiltration, fibrous tissue hyperplasia, synovial cell hyperplasia and macrophage hyperplasia of the synovial tissues were observed. Results The rating score and total score in terms of the synovial cell hyperplasia(P〈0.001), inflammatory cell infiltration (P〈0.01), macrophage hyperplasia (P〈0.01) and fibrous tissue hyperplasia (P〈0.001) of the model group were significantly reduced, compared with the normal group. After treatment, the rating score and total score of treatment group showed different degrees of reduction in the above-mentioned aspects, of which, the score for the fibrous tissue hyperplasia of the 50 mW laser acupuncture group (P〈0.01), the score for the synovial cell hyperplasia (P〈0.05) and fibrous tissue hyperplasia (P〈0.01) of the 100mW laser acupuncture group as well as the total score (P〈0.01) of the 150mW laser acupuncture group were significantly better than that of the model group. In addition, the score for the synovial cell hyperplasia of the 100mW laser acupuncture group was significantly better than that of the drug group (P〈0.05). Conclusion The laser acupuncture has significant inhibition effect on the pathological changes such as synovial tissue hyperplasia, fibrosis hyperplasia of CIA rat, and there is certain relationship between the effects and the parameter setting of laser.
