胞内高迁移率族蛋白B1对高糖诱导的小鼠心脏成纤维细胞转化生长因子-β1表达的影响 被引量:1

摘要 目的:探讨胞内高迁移率族蛋白B1(HMGB1)在高糖诱导的小鼠心脏成纤维细胞表达转化生长因子-β1(TGF-β1)中的作用。方法:酶消化法分离3周C57/BL6小鼠心脏成纤维细胞,培养并传代,实验使用2~4代细胞,波形蛋白(vimentin)免疫细胞化学染色鉴定细胞。高糖培养不同时间(0,6,12,24,48h)后分别检测成纤维细胞HMGB1、TGF-β1 mRNA的表达量。在转染siRNA后再高糖培养心脏成纤维细胞6 h,RT-PCR检测HMGB1、TGF-β1 mRNA的水平。结果:与0h相比,高糖培养6,12,24,48 h后成纤维细胞HMGB1、TGF-β1 mRNA的表达都上调。转染HMGB1 siRNA后高糖培养6 h的成纤维细胞TGF-β1表达下调。结论:HMGB1可能参与调节高糖诱导的心脏成纤维细胞的TGF-β1的表达。 Objective: To investigate the effect of HMGB1 on the expression of TGF-β1 of cadiac fibroblast induced by high glucose.Methods: The cardiac fibroblasts were isolated from 3 w C57/BL6 mice.The identification of cardiac fibroblasts was evaluated by immunocytochemical staining with vimentin.The cardiac fibroblasts were treated by high glucose for different times(0,6,12,24,48 h).After transfected with siRNA,the cardiac fibroblasts were treated by high glucose for 6h.The mRNA levels of HMGB1 and TGF-β1 of cardiac fibroblasts were determinated by RT-PCR.Results: The cultured cells were in accordance with characteristics of cardiac fibrolast.The mRNA levels of TGF-β1 of cardiac fibroblasts were inhibited by HMGB1siRNA after treated by high glucose for 6 h.Conclusion: HMGB1 may contribute to modulate the expression of TGF-β1 of cardiac fibroblasts induced by high glucose.
