参麦注射液对大鼠肢体缺血-再灌注损伤后骨骼肌细胞凋亡的影响 被引量:1

Effects of Shenmai Injection on Skeletal Muscle Apoptosis Following Limb Ischemia -reperfusion Injury in Rats

摘要 目的:探讨参麦注射液对大鼠肢体缺血再灌注损伤的保护机制。方法:30只SD大鼠,随机分为假手术组、模型组和参麦组,每组10只。模型组与参麦组采用扎闭股动、静脉缺血4h,开放再灌注1h制造缺血再灌注模型,并分别于再灌注即刻给予生理盐水或参麦注射液4mL/kg腹腔注射;假手术组不结扎股动静脉,4h后给予生理盐水4mL/kg腹腔注射。采用免疫组织化学方法检测腓肠肌组织Bcl-2及Fas蛋白表达情况。结果:与模型组比较,参麦组腓肠肌组织Fas蛋白表达明显降低(P<0.05),Bcl-2蛋白表达增加(P<0.05)。结论:参麦注射液可减轻大鼠缺血再灌注后骨骼肌细胞凋亡,对缺血再灌注损伤有一定的防治作用。 Objective:To investigate the protective effect of Shenmai injection on skeletal muscle cell apoptosis fol- lowing limb ischemia-reperfusion injury in rats. Methods:Thirty SD rats were randomly divided into three groups (n=10),including sham-surgery group,ischemic reperfusion+normal saline group (I/R group),and ischemic reperfu- sion+Shenmai injection group(SM group). Ischemic reperfusion models were made by femoral vessels ligation for 4h and reperfusion for lh. I/R group and SM group were given saline or Shenmai injection of 4 ml/kg at the very be- ginning of reperfusion. Ischemia-reperfusion injury was not induced in the sham-surgery group (S group). Immuno- histochemistry was used to detect the expressions of Bcl-2 and Fas in gastrocnemius tissues. Results:Compared with the I/R group,the Fas in gastrocnemius tissues was significantly lower(P〈0.05),but the Bcl-2 was significantly higher in the SM group (P〈0.01). Conclusion:Shenmai injection can inhibit skeletal muscle cell apoptosis and protect skeletal muscle cells from ischemia-reperfusion injury
