摘要
目的探讨中西医结合方法逆转人结肠癌耐药株多药耐药的机制。方法用四甲基偶氮唑蓝(MTT)法测定5-氟尿嘧啶(5-Fu)对实验组及对照组细胞的细胞毒作用,测定其半数抑制浓度,计算其逆转倍数(RI),并用流式细胞仪测定细胞内罗丹明的滞留浓度,分析P-糖蛋白的滞留浓度。结果中西医结合法可以明显增加化学治疗药物对结肠癌耐药细胞的杀伤作用(P<0.05),逆转了其多药耐药作用,逆转指数为2.92,并减少了P-糖蛋白对化学治疗药物的外排。结论中西医结合法对结肠癌多药耐药逆转效果明显,其机制可能与其中药成分抑制P-糖蛋白的外排作用有关,值得进一步研究开发。
Objective To investigate the combined method of reversing human colon cancer resistant strains muhidrug resistance mechanism of traditional Chinese medicine and Western medicine. Methods To determine the 5-FU of the experimental group and the control group cell cytotoxicity by MTT method, determination of the half inhibitory concentration, the reversal index calculation, and determination of intracellular Rhodamine flow cytometry retention con- centration, namely stranded concentration P-glycoprotein. Results The traditional Chinese and Western medicine can significantly increased the killing effect of chemotherapy drugs on colon cancer drug-resistant cell (P〈0.05), reversed the multi-drug resistance, reversal index was 2.92, and could reduce the P glycoprotein on the efflux of chemotherapeutic drugs. Conclusion Traditional Chinese and Western Medicine on colon cancer muhidrug resistance reversal effect, its mechanism may be associated with traditional Chinese medicine compositions to inhibit P-glycoprotein efflux effect, worthy of further research and development.
