高糖通过高渗透压改变PAX3基因表达抑制RSC96雪旺细胞增殖 被引量:4

摘要 探讨高糖对外周神经雪旺细胞增殖的影响,寻找糖尿病神经病变发病新机制.应用CFSE染色经流式细胞仪及CCK8试剂盒分析RSC96雪旺细胞在高糖作用下的增殖情况,采用RTPCR技术及westernblot检测PAX3基因及蛋白表达.结果显示,高糖及甘露醇均能抑制雪旺细胞增殖;其对应的增殖相关基因PAX3及蛋白表达下调.高糖可通过高渗透压抑制雪旺细胞增殖,可能与PAX3基因有关,这一结果将为糖尿病神经病变损伤后修复的研究提供一个新的研究靶点. The influence of high glucose on proliferation of Schwann cells is studied in this paper. The flow cyto-metric fluorescence of CFSE and CCK8 ( cell counting kit - 8 ) are employed to analyze the proliferation of RSC96 Schwann cells. Total mRNA and protein expression of PAX3 gene are then detected by RT - PCR and western blot. It is shown through the results that both high glucose and mannitol inhibit proliferation of RSC96 Schwann cells and that the gene and protein of PAX3 is down - regulated. High glucose through osmotic pressure inhibits proliferation of RSC96 Schwann cells, the mechanism of which may be related to down - regulation of PAX3 gene. The study will provide a new investigated target for diabetic peripheral neuropathy.
