摘要
目的:研究在葡萄胎恶变过程中外周血CD4+CD25+调节性T细胞(Treg)所起的作用。方法:收集27例葡萄胎患者作为观察组(A组,含轻中重度各9例),以15例恶变葡萄胎患者作为阳性对照(B组,含侵袭性葡萄胎7例及绒癌8例),并以15例同龄段健康女性作为阴性对照(C组)。采用流式细胞术检测上述三组外周血Treg及其功能因子白介素(IL-10)、转化生长因子β1(TGF-β1)及叉头蛋白p3(Fox p3)的百分含量。结果:A组Treg含量(8.79±3.74)%,B组(13.28±5.43)%,C组为(4.29±1.33)%,以上三组当中任意两组间Treg含量差异均具有显著性(P=0.012)。三组间TGF-β1及Foxp3的变化趋势与Treg一致;而A组患者IL-10与C组无显著性差异,但A组患者IL-10显著低于B组(P<0.05)。轻度葡萄胎患者的Treg及TGF-β1百分含量均显著低于中度及重度组(P<0.05),且重度患者的Fox p3显著高于轻度及中度(P<0.05)。结论:CD4+CD25+调节性T细胞及其胞内因子TGF-β1及Fox p3可能参与葡萄胎恶变的发生,且与葡萄胎的严重程度相关。
Objective:To investigate the process of hydatidiform mole malignant peripheral blood CD4+CD25+ regulatory T cells(Treg) role.Methods:collection of 27 cases of hydatidiform mole patients as observation group(group A,including light relation between all the 9 cases),15 cases of patients with malignant hydatidiform mole as positive control(group B,including 7 cases with invasive hydatidiform mole and velvet cancer 8 cases),and 15 patients age period of healthy women as negative control(group C).Using flow cytometry to detect the three sets of peripheral blood and Treg function factor,interleukin(IL-10),transforming growth factor β1(TGF-β1) and fork protein p3(Fox p3) percentage.Results:group A Treg content is(8.79±3.74)%,(13.28±5.43)% for group B,and C group is(4.29±1.33)%,any of the above three groups Treg content between the two groups have significant difference(P=0.012).Between the three groups of TGF-β1 and the change trend of Fox p3 and Treg consistent;And IL-10 patients of group A and group C there was no significant difference,but IL-10 patients of group A was significantly lower than group B(P0.05).Mild hydatidiform mole patient Treg and TGF-β1 percentage were significantly low moderate and severe group(P0.05),and the heavy Fox p3 significantly higher in patients with mild and moderate(P0.05).Conclusion:CD4+CD25+ regulatory T cells and intracellular factors TGF-β1 and Fox p3 may participate in hydatidiform mole of malignant transformation,and is associated with the severity of hydatidiform mole.
