富硒甘蓝提取物对肺癌细胞NCI-H446生长抑制的研究 被引量:2

摘要 采用气相色谱-质谱联用技术测定富硒甘蓝中硒的存在形式,并用富硒甘蓝提取液对人肺癌H446细胞进行肿瘤生长抑制试验.结果表明,富硒甘蓝中有机硒主要以甲基硒半胱氨酸和硒代蛋氨酸的形式存在,另有痕量的硒半胱氨酸;此提取物能显著抑制H446细胞的生长,并呈时间和剂量依赖效应.细胞形态观察发现,在硒质量浓度高于2mg·L-1时即可诱导H466细胞皱缩,表面微绒毛减少和凋亡小体的出现,说明以硒甲基硒半胱氨酸为主要富硒形式的甘蓝,可能通过诱导细胞凋亡抑制肿瘤细胞生长. We analyzed selenium forms in selenium enriched cabbage by gas chromatography-mass spectrometry, and then investigated the inhibiting effects of selenium-enriched cabbage extract on hu- man lung cancer H446 cells. The results showed that the organic selenium exists in cabbage mainly in the form of methylselenocysteine and selenomethionine with the trace of selenocysteine. Cabbage ex- tract can significantly inhibit the growth of H446 cells in a time- and dose-dependent models. Cell mor- phology observation found that selenium concentration higher than 2 mg ·L^-1could induce H466 cell shrinkage, reduce surface microvilli, and result in cell apoptotic.
