猕猴桃多糖对615小鼠胃癌原位移植瘤PCNA、p53表达的影响 被引量:9

Influence of actinidia chinensis polysaccharide on expression of PCNA and p53 in orthotopic transplanted cancer of gastric tumor in 615 mice

摘要 目的:研究猕猴桃多糖对615小鼠胃癌原位移植瘤的抑瘤效应及其与增殖细胞核抗原(PCNA)、p53表达的关系。方法:用OB吻合胶粘贴法建立615小鼠胃癌原位移植瘤模型。40只615小鼠随机分为5组:猕猴桃多糖高、中、低剂量组、5-FU对照组、模型对照组。计算抑瘤率、脾指数,并用免疫组织化学方法检测PCNA、p53的表达。结果:与模型对照组比较,用药各组平均瘤重明显下降(P<0.01);猕猴桃多糖中、高剂量组的脾指数明显升高(P<0.05,P<0.01);用药各组PCNA、p53的平均阳性指数有显著下调(P<0.01)。结论:猕猴桃多糖对615小鼠胃癌原位移植瘤具有抑制作用,且随着剂量的增加其抗肿瘤作用增强。其作用途径可能与提高小鼠的免疫功能及下凋PCNA、p53的表达有关。 Objective: To investigate the effects of actinidia chinensis polysaccharide inhibiting orthotopic transplanted cancer of gastric tumor in 615 mice and the relationship between anti-cancer effects of actinidia cbinensis polysaccharide and the expression of PCNA and p53. Methods: Models of orthotopic transplanted cancer of gastric tumor in 615 mice were established with OB anastomosis adhesive stick method. 615 mice were randomly divided into 5 groups: actinidia chinensis polysaccharide high, medium and low dose group, 5-FU positive control group, model control group. The anti-cancer rate and the spleen index was calculated, and the expression of PCNA and p53 was detected by immunohistochemical methods. Results: Compared with model control group, the average tumor weight of medication group decreased significantly, there was significant differences (P〈0.01); the average spleen index of the actinidia chinensis polysaccharide in medium and high dose groups increased significantly, it showed significant differences (P〈0.05, P〈0.01); the average positive index of PCNA and p53 of dosage groups were significantly lower (P〈0.01). Conclusion: Actinidia chinensis polysaccharide can inhibit the growth of orthotopic transplanted cancer of gastric tumor in 615 mice, the more dosage, the better effect its mechanism may partly be to improve the immune function in mice and to lower the expression of PCNA and p53.
