阴虚、阳虚体质血清对肺癌细胞NF-κB、P16基因表达的影响 被引量:7

Effects of yin-yang deficiency constitutional types serum on NF-κB and P16 expression in lung cancer cell

摘要 目的:探讨阴虚、阳虚体质血清对肺癌细胞生长的部分机制。方法:肺癌细胞A549培养使用10%的人血清,采用MTT分析细胞增殖,流式细胞仪分析细胞周期,用TR-PCR及Western Blot的方法测定基因表达。结果:阴虚、阳虚体质血清培养的细胞增殖数明显高于平和质(P<0.05),G_0/G_1期的细胞数低于平和质(P<0.05),而S期的细胞数明显高于平和质(P<0.05),而且阴虚、阳虚体质血清培养的细胞内表达的核转录因子-κB(NF-κB)高于平和质(P<0.05,P<0.01),P16则低于平和质(P<0.05)。结论:阴虚、阳虚体质血清能促肺癌细胞增殖,可能的机制是促进细胞内NF-κB基因表达,而抑制P16基因表达有关。 Objective: To investigate mechanism of yin-yang deficiency constitution types serum on lung cancer cell growth. Methods: A549 cell culture technique was treated with 10% human serum. Cell proliferation was observed by MTT metabolism measuring. Flow cytometry was performed to track cell cycle progression. Western bolts and RT-PCR were performed to evaluate gene expression. Results: The number of cell proliferation of yin-yang deficiency constitutional types serum was significantly higher than that of gentleness type senum (P〈0.05). Flow cytometry (FCM) analysis indicated that the GJG~ phase fraction ratio of yin deficiency, yang deficiency serum group was lower than that of serum of gentleness type (P〈0.05), while the S-phase fraction ratio was higher than that of serum of gentleness type (P〈0.05). Western blots and RT-PCR results displayed that serum of constitution of yin deficiency, yang-deficiency elevated the expression of NF-~cB (P〈0.05, P〈0.01) and decreased the expression of P16 than serum of gentleness type did (P〈0.05). Conclusion: These results suggest that serum of constitution of yin deficiency, yang deficiency significantly sitimulate cell proliferation. This effects may be related to increasing the NF-~cB expression and decreasing P16 expression.
