摘要
目的:观察参附益心颗粒对心衰大鼠心肌血管紧张素Ⅱ1型受体和2型受体(AT1和AT2)表达的影响。方法:随机将150只雄性SD大鼠分为造模组130只与假手术组20只,结扎大鼠左冠状动脉前降支(LAD)建立心衰大鼠模型,假手术组手术方法同上,仅在LAD下穿线不结扎。造模4周后检测左心室射血分数(LVEF)<50%者随机分为5组,每组10只,分别为卡托普利组、氯沙坦组、参附益心颗粒高、低剂量组、模型组,另选假手术组大鼠10只。假手术组及模型组以纯净水灌胃,药物干预组以药物溶液灌胃给药,每天1次,4周后查LVEF、称重后取标本,免疫组化法检测大鼠心肌AT1、AT2表达。结果:参附益心颗粒能明显提高心衰大鼠LVEF。心衰大鼠AT1、AT2表达显著增强,参附益心颗粒可明显降低AT1、AT2表达。结论:参附益心颗粒可改善心衰大鼠心功能,下调心肌中AT1、AT2表达水平阻断肾素血管紧张素系统,可能会起到延缓或改善心肌重塑的作用。
Objective: To study the effect of Shenfuyixin Granule on the expression of myocardial angiotensin 1I type 1 receptor and type 2 receptor (AT1 and AT2) in heart failure rats. Methods: 150 heathly male SD rats were randomly divided into sham-operated group (n=20) and model group (n=130). In model group the left anterior descending coronary artery was occluded, in sham operation group the sutures were passed under the left anterior descending coronary artery without ligation. After 4 weeks the modeling group were randomly assigned to 5 groups (n=10) based on the value of LVEF〈50% obtained by Color Doppler Ultrasound, such as captopril group, losartan group, two groups of Shenfuyixin Granule (low dose, high dose), model group. All the medicine was given through gavage once a day for 4 weeks, the rats in model group and sham operation group (n=10) were administered gavage with equivalent sodium chloride once a day for 4 weeks. After 4 weeks The LVEF was valued again with Color Doppler Ultrasound; the tissue was collected to detect index. Results: The LVEF was improved markedly in Shenfuyixin Granule. Two Shenfuyixin Granule groups obviously decreased myocardial AT1 and AT2. Conclusion: Shenfuyixin Granule can improve Cardiac function of CHF rats, decrease myocardial AT1 and AT2 to block RAS, maybe delay or improve cardiac remodeling.
