摘要
目的:观察消癥散对人肝癌HepG-2细胞凋亡的影响,探讨其抗肿瘤的可能作用机制。方法:MTT比色法检测消癥散对人肝癌HepG-2细胞的生长抑制作用;倒置显微镜观察细胞形态学的改变;RT-PCR法检测消癥散对人肝癌HepG-2细胞p53和Bcl-2 mRNA表达的影响。结果:消癥散对人肝癌细胞HepG-2的生长活性有抑制作用,并可调节人肝癌HepG-2细胞中p53和Bcl-2 mRNA的表达水平。结论:消癥散能促进人肝癌HepG-2细胞凋亡,其作用机制可能与下调Bcl-2和升高p53相关。
Objective: To observe the effect of Xiaozhen San on apoptosis of human hepatoma HepG-2 cells and study the mechanism of action. Methods: Inhibitory effect of Xiaozhen San on the proliferation of HepG-2 cells was tested by MTF assay, and the morphology changes were observed by inverted microscope assay. RT-PCR was used to examine the effect of Xiaozhen San on p53 and Bcl-2 mRNA expression in HepG-2 cells. Results: Proliferation of HepG-2 cells was markedly inhibited by Xiaozhen San, and Xiaozhen San could inhibit the expression of Bcl-2 and increase expression of p53. Conclusion: Xiaozhen San can induce apoptosis of HepG-2 cells, its mechanisms might be probably involved in the down-regulation of Bcl-2 expression and the activation of p53 in this process.
