复方健耳剂对国产DBA/2J小鼠耳蜗组织脑源性神经营养因子表达影响的初步研究 被引量:4

Preliminary study of the impact from Chinese herbal Compound Jian-er Agent on BDNF expression in cochlear tissue of the domestic DBA/2J mice

摘要 目的:研究中药复方健耳剂对小鼠耳蜗组织脑源性神经营养因子(BDNF)表达的影响,探讨其保护老年性耳蜗感音神经性损害机制。方法:选择断奶DBA/2J小鼠10只,随机分为对照组和中药组,各5只,对照组小鼠为常规饲料与日常饮水喂养;中药组小鼠为常规饲料与中药溶液喂养;至105d龄时两组同时终止实验,取出耳蜗,利用Real-Time PCR技术定量检测两组耳蜗组织BDNF的表达,并统计分析。结果:中药组BDNF的表达比对照组高(P<0.01)。结论:复方健耳剂对BDNF的表达影响可能是其有效对抗老年性耳蜗感音神经性损害的重要机制之一。 Objective: To study the impact from Chinese herbal compound Jian-er agent on BDNF expression in cochlear tissue of mouse, and explore the mechanisms of their preventing age-related cochlear sensorineural damage. Methods: 10 DBA/2J mice after ablactation were randomly divided into 2 groups, with 5 animals in control group, 5 in experimental group of Chinese herbal compound (CHC). The animals in control group were fed regularly, while experimental group of CHC were fed regularly and commonly fed water was substituted by CHC solution. Until the 105 days of experimental course, the experiment was ended for all the 2 groups of animals at the same time. We took cochlea from intracranial and detected quantitatively cochlear BDNF expression in the cochlear tissue by using Real-Time PCR technology, and made a statistical analysis. Results: The amount of BDNF expression in experimental group of CHC were higher than that in control group (P〈0.01). Conclusion: The impact of CHC on BDNF expression may be one of the important mechanisms of CHC effectively preventing age-related cochlear sensorineural damage.
