摘要 目的:探究干酪乳杆菌胞外多糖(EPS)对体外培养的小鼠腹腔来源巨噬细胞分泌炎症相关因子的影响。方法:采用倒置显微镜,观察干酪乳杆菌EPS对腹腔来源巨噬细胞形态的影响;采用双抗体夹心ELISA和Griess法,分析干酪乳杆菌EPS对腹腔来源巨噬细胞分泌促炎症因子TNF-α、NO及抗炎症因子IL-10的变化。结果:EPS能够抑制由脂多糖导致的巨噬细胞形态的变化,与空白组相比,350、400和450μg/ml EPS均能提高TNF-α、NO及IL-10的分泌量。脂多糖(Lipopolysac-charide,LPS)能诱导TNF-α和NO的分泌,抑制IL-10的分泌。350、400和450μg/ml EPS能抑制由10μg/ml脂多糖诱导的TNF-α和NO的分泌,促进LPS抑制的巨噬细胞分泌IL-10,且呈剂量依赖关系。结论:干酪乳杆菌EPS能双向调节体外培养的腹腔巨噬细胞分泌炎症相关因子功能。 Objective:To explore the effect of Lactobacillus casei exopolysaccharide(EPS) on inflammation-related factors secretion of macrophages in the mouse peritoneal in vitro.Methods: Using an inverted phase microscop,we observed the morphology of macrophages in the mouse peritoneal exposed to Lactobacillus casei EPS.The level of mouse TNF-α and IL-10 in culture supernatant was determined by sandwich enzyme-linked immunosorbent assay.The level of NO was detected by NO analyzing Kit.Results: EPS decreased the LPS-induced morphological changes and their secretion of the proinflammatory factor TNF-α,NO and anti-inflammatory factor IL-10 were increasing after treatment with 350 μg/ml,400 μg/ml and 450 μg/ml EPS compared with untreated controls.Lipopolysaccharides(LPS) induced the secretion of TNF-α,NO and suppressed the secretion of IL-10 by peritoneal macrophages.350 μg/ml,400 μg/ml and 450 μg/ml EPS suppressed 10 μg/ml LPS-induced release of TNF-α and NO by peritoneal macrophages and promoted 10 μg/ml LPS-induced suppression of IL-10,with dose-dependent effect.Conclusion: The EPS of Lactobacillus casei could influence inflammation-related factors secretion function of peritoneal macrophages by two-way regulation.
